Abstract: The purpose of this thesis is to report quantum chemical investigations into a series of problems related to contemporary organometallic chemistry. In recent years density functional approaches have become the most commonly used theoretical methodology in organometallic chemistry. Alongside these developments, a number of new tools for analyzing electronic structure such as topological analysis of electron density (Atoms in Molecules theory) have emerged. As a result, theory is now in a position to compute ab initio many of the experimental observables that underpin modern organometallic chemistry. The bonding in the Triosmium carbonyl cluster 1 [Os3(? - H)(? - ?2 - dpa - N,N)(CO)10] and Triosmium carbonyl cluster 2 [Os3( ? - H)2(NHC)(CO)9] are explored using the Quantum Theory of Atoms - in - Molecules (QTAIM). The metal - metal and metal - ligand bond critical points properties ?(r), ?2?(r), H(r), G(r), V(r) and ellipticity, and also the bond delocalization indices ?(A, B), are correlated with the data from previous studies of the organometallic systems. These results have allowed a comparison between topological properties of different atom - atom interactions. In the core of dihydride triosmium cluster 2, Os3H2 part, the topological data recognizes the existence of a bond path in the dihydride bridged Os atoms, Os(1) - Os(3) edges, whereas in monohydride triosmium cluster 1 there is no direct bond path has been found for the interaction between the hydride bridged Os atoms, although a non - negligible delocalization index ?(Os(1)...Os(2)) has been obtained for this non - bonding interaction. A multicenter 4c - 4e interaction is proposed to exist in the core part, Os3H, in cluster 1 and bridged part, Os(1) - H(1) - Os(3) - (H2) in cluster 2. In addition, an interaction of 5c - 5e type is proposed to exist in the core part, Os3H2. All topological parameters calculated for the Os - N and Os - C bonds between the osmium atoms and the pyridyl and NHC ligands in compound 1 and 2, respectively, are similar, and they confirm that these interactions are pure ? bond. The analysis of the topological parameters of the NHC and pyridyl ligands bonds confirm the existence of ? - electron delocalization within the six - membered ring of pyridyl ligand and hindered ? - electron delocalization within the five - membered ring of NHC ligand with some double - bond character in the interaction of the carbine C atom with the adjacent N atoms.

Abstract: تضمن الجزء الاول من البحث تحضير اربعة كواشف عضوية جديدة هي مشتقات ازو لمركب الباراكريزول وقد تم تشخيص الكواشف المحضرة من خلال اطياف الاشعة فوق البنفسجية - المرئية (UV - Vis) فضلا عن قياس اطياف الاشعة تحت الحمراء IR)) وتم اجراء قياسات نسب العناصر الدقيق ( | The first part of this study included perpetrating four new organic reagents which are derivatives of P - cresol. Its identification was done by UV - Vis spectrophotometer, IR and C. H. N. Depending on the results, we found the correspondence between the theoretical calculation and the experimental ones with a way to suggest the final structures of the prepared reagents. The purity of these reagents was tested through thin layer chromatography. The prepared reagents are : 1. 2[(4 - Methyl phenyl )azo] - 4 - Methyl phenol (MePAMP)2. 2[(4 - Chloro phenyl )azo] - 4 - Methyl phenol (CPAMP)3. 2[(4 - Methoxy phenyl )azo] - 4 - Methyl phenol (OMePAMP)4. 2[(4 - Nitro phenyl )azo] - 4 - Methyl phenol (NPAMP)The second part deals with the study of chromatographic behavior of organic prepared reagents dissolved in ethanol according to gas chromatography on stationary liquid phases as capillary columns with different polarities as zebron (FFAT) which is characterized with high polarity, silica fused (BP10) with a medium polarity and poly phenyl siloxine (OV - 5) that appears with low polarity.Gas chromatography was equipped with a flame ionization detector (FID) and the nitrogen as carrier gas with flow rate 30cm3min - 1 at different column temperatures ranged 100C increments depending on columns maximum operation temperature, column inlet temperature and detector temperature were higher than the separation column (25,50) 0C respectively. This investigation attempts to find the optimum conditions for the sequential separation were defined for the azo compounds of all capillary columns such as injection, applied pressure inlet of column, gas flow rate as well as the conditions of the efficient separation. The order of elution and resolution of compounds on various stationary liquid phases were normal chromatographic behavior which is decrease in specific retention volumes with increase of column temperature through the relationship between logarithm of specific retention volume for the studied compounds against reciprocal of absolute column temperature which is observed generally linear relation for all compounds on different liquid phases by using (Clausius - Clapeyron) , then calculate the thermodynamic parameters ?H0 , ?S0 and ?G from the slope and intercept respectively the negative values of partial molar enthalpy ?H0 of solutions indicate to highest interaction between liquid phases and separated compounds. The results show that the zebron (FFAT) consider the high negative value of ?H and the reaction was exothermic, on other hand the highest negative value of ?s means less random of solutes on some liquid phase , so it was more selective towards to prepared azo derivatives of p - cresol. Also calculated separation factor (RS),number of theoretical plate (N), equivalent highest of one theoretical plate (HETP) respectively for mixture of studied compounds on various liquid phases. The precision and accuracy of the prepared organic reagent (MePAMP) from 0.06% to 1.44% for (CPAMP) from 0.91% to 1.5%, (OMePAMP) from 0.03% to 1.42% and finally for (NPAMP) from 0.06% to 1.6%

وفي هذا الصدد، نعتقد ان العمل البحثي الحالي سوف يسهم في فتح افاق جديدة في تصم | The separation and extraction of trace amounts of medicaments in biological specimens, as well as in pharmaceutical formulations have not received much attention by using the so - called cloud - point extraction (CPE). In this context, we think that the present research work will contribute to opening new prospects in designing procedural steps and expands the applications of the cloud point extraction methodology in most important areas, including pharmaceutical sciences, forensic and environmental analyses. The present work concentrates on the developing of new eco - friendly procedures for determination of two selected antibiotic medicaments namely cirprofloxacin chloride (CIPRO) and norfloxacin (NOR) using iron (III) ion in human serums and pharmaceutical formulations, as well as the exploitation of these two drugs in chemical analysis as chelating agents for the detection of iron in pharmaceutical formulations via using cloud point extraction coupled with molecular spectrophometry. Generally, these methods implicate the use of a nonionic surfactant (Triton X - 114) as an extracting medium which entrap the hydrophobic colored complex formed between one of these drugs and iron (III) ion in acidic medium as a reaction system for designing the CPE procedures. This thesis includes three main chapters as follows;Chapter One summarizes some theoretical and practical principles of cloud point extraction (CPE) methods and then highlights the knowledge of the target analytes (CIPRO, NOR and iron) in term of their structures, pharmacological importance besides a concise chemical literature review for the estimation of these analytes by different analytical techniques. Chapter Two consists of an outline of instrumental techniques , general apparatus and chemicals used in the present work. The full analytical procedures for CPE which have been designed for the determination of the target analytes under study are made in this thesis.Chapter Three covers a detailed study for the discussion of the analytical data obtained throughout this work which can be summed up as follows;A. Formation of chelates between CIPRO or NOR and Fe(III) in acidic medium , where the drugs have individually been estimated in the resulting complex by spectrophotometry after the cloud point extraction at each respective ?max, and the use of developed method so that to determine any of the interested drug in human serum and pharmaceuticals. At first, the study was conducted to pinpoint the optimum conditions for the formation of complexes by CPE and the results were as follows; 1. The optimum experimental conditions for Fe - CIPRO and Fe - NOR complex for the determination of CIPRO and NOR respectively : H2SO4 concentration (5x10 - 4 M and 2.5x10 - 4M; Fe(III) concentration ( 6 ?g mL - 1 and 10 ?g mL - 1); Triton X - 114 amount ( 1%) for both drugs; temperature and time ( 75 °C at 25 min and 65 °C at 20 min).2. The achieved analytical figures of merit on applying the developed methods for CIPRO and NOR are : linear range (2.5 - 120 ?g mL - 1 ) for drugs, limit of detection (0.770 and 0.204 ?g mL - 1), Enrichment factor (143 and 280 fold), extraction efficiency (96.46 and 96.62%) and mean recovery percentage (98.89±0.87% and 98.95±1.09). 3. The newly established procedures have been applied for the determination of CIPRO and NOR in human blood serum and pharmaceuticals. For the determination of these medicaments in serum samples , the statistical paired t - test was used to test the significance of the proposed method comparing with the conventional UV - spectrophotometry, while in pharmaceuticals, the findings were compared statistically with quoted values that stated by the manufactures. B. Iron (III) ion was also determined in the pharmaceutical formulations based on the above reaction system in (A) via formation of yellow colored complexes with each medicament which extracted by micelles generated by CPE and subsequently detected spectrophotometrically at the same ?max of the above mentioned complexes. The optimum established conditions and the analytical data obtained could be summarized as follows : 1. The same optimum conditions were used in (A) except that the concentration of CIPRO or NOR here was of 7x10 - 5 M and Triton X - 114 amount of 0.6% in final solutions.2. The analytical figures of merit obtained on applying the developed methods for iron using CIPRO and NOR respectively; linear range (5 - 150 ng mL - 1); limit of detection (2.67 and 3.42 ng mL - 1); preconcentration factor (71 and 83 fold); mean recovery percentage (99.78±0.53% and 98.77±2.29); extraction efficiency (99 and 98.7%). 3. The effect of some additives of the pharmaceutical formulations was also studied and the iron determined in three selected formulations collected from local drugstores. The results were compared statistically with quoted values and shown not to be significant at 95% confidence level.

Abstract: There are a wide variety of polyphenolic compounds named flavonids. These are extensively distributed in plants, and they are considered to be dietary antioxidants. They occur naturally in broad range of vegetables, fruits, and primarily recognized as the pigments. Many flavonoids have a metallic ion complexation capacity. The study of this capacity is important because it can be used in producing nutritional supplements, medicine, and heavy metal detoxification.Chelation therapy is the use of chelating agents (complexing agents) to detoxify poisonous metal such as mercury, arsenic, and lead by converting them to chemically inert form that can be excreted without further interaction with the body.Lead is considered as one of the most hazards and cumulative environmental pollutants that affect all biological system through exposure from air, water and food sources. It is toxic heavy metals which are known to induce a broad range of physiological, biochemical and behavioral dysfunction in humans. Because of its perverseness in the environment, lead poisoning still remains an important health problem. The polarographic measurements were carried out using differential pulse polarography (DPP), at hanging mercury drop electrode (HMDE) at the optimal conditions for flavonoids which were 1M HCl for Quercetin, Rutin, and Hespertin and 0.01M NaOH for chrysin as a supporting electrolyte.A well - defined two electron reversible reduction and diffusion controlled one wave for these flavonoide were observed, a linear relationship between concentration and diffusion current (id) was obtained with the value of correlation coefficient (r2) near to one, this obey Ilkovic equation. The values of n; the number of electrons transferred in the reduction process were obtained from the slop of the straight line corresponding to E vs. log i/id - i.Addition of flavonoides to a solution of Pb (II) ion, show the appearance of a new peak in a more negative potential than Pb (II) peak, with the gradual decrease in id of Pb (II) ion which suggests the complex formation between them. The stoichiometry and the stability constant of Pb (II) complex with flavonoids were calculated by Lingane equation; a plot of ?Ep vs. log [L] give a straight line, the slope of the line allows the determination of the coordination number (p), and the stability constant were determined from the intercept, the stoichiometry of these complexes were (1 : 1) with a stability constant of : Pb - Quercetine (5.5*107) ? Pb - Rutine (1.3*107) ? Pb - Hespertine (4*106) ? Pb - Chrysin (5.4*104)These measurements also includes the determination of the kinetic parameter of the electrode reaction (Kth, ?n) and the thermodynamic parameters such as ?H, ?G, and ?S.The spectrophotometric measurements were carried out in 50% methanol/water for Quercetin and Rutin, 30% methanol/water for Hespertin and in phosphate buffer of pH - 7 for Chrysin. The electronic spectrum of free flavonoides in methanol solution was mainly characterized by an important absorption band at ? 370 nm (band I) and a second one located at ? 256 nm (band II). Upon addition of lead (II) to the flavonoids solution significant changes are observed in the electronic spectra, a bathochromic shift in band (I) to the extent of ? 40 nm and a shift in band (II) to the extent of ? 7 nm, the change in ?max and the change in the intensities of the bands were due to the coordination between flavonoids molecules and Pb (II) ion.The stoichiometry of the formed complexes were investigated by the method of continuous variation and it were found (1 : 1), the stability constant were calculated also and were found to be in the order of ? 105 to 106.Thermodynamic parameters and kinetic studies were also determined which indicates a second order interaction between lead (II) and these flavonoides.Spectroscopic measurements for (EDTA) Complexation with lead (II) were carried out in order to compare its stability constant with that obtained for flavonoides complexes, due to its application in the treatment of metal poisoning.Stability constant of (EDTA) with Pb (II) complex were in the order of 108 which is much more efficient than flavonoides.

The number of scabietic patients who attended the outpatient clinic during February to November 2012 were 300 (168 males and 132 females). Their ages ranged from 10 to 65 years. They were included to show the prevalence of scabies. Sixty scabietic patients (out of 300 patients) who did not have allergic diseases, helminthic infections, previous attack with scabies, and/or getting any antihistamines drugs were included for human IL - 1?, TNF - ?, IL - 4, IL - 5 and total IgE assays using ELISA technique; superoxide dismutase (SOD) activity assay and eosinophils(%). These assays were applied on scabietic patients (who were divided according to onset of symptoms or duration of scabies symptoms into early and late scabietic patients) as well as healthy controls. Also Sarcoptes scabiei mite proteins were extracted. Then heat stable mite proteins concentration was determined by Bradford’s method. SDS - Polyacrylamide Gel Electrophoresis (SDS - PAGE) analysis was used. The activity of mite allergens was assayed by skin prick test (SPT) in 10 scabietic patients and 5 controls with 1.2 ?g/ml and 2.4 ?g/ml. House dust mite (HDM) antigens were skin prick tested in patients with scabies and controls. The prevalence of scabies in current study constituted 6.54% from all the dermatoses which attended the clinic. The males represented 56% and females were 44%. The age group (10 - 19) years was the highest prevalence of scabies (41.7%). Early scabietic patients represented 72% versus late scabietic patients were 28%. The results revealed that a highly significant elevation (p< 0.001) in serum cytokines (IL - 1?, TNF - ?, IL - 4 and IL - 5) as well as IgE, SOD and eosinophils (%) in the two groups of scabietic patients in comparison to the controls. Results of correlation demonstrated that IgE, SOD and eosinophils (%) were positively and significantly correlated (p< 0.001) with the studied cytokines in early, late scabietic and control groups. IgE was positively correlated with IL - 1?, TNF - ?, IL - 4 and IL - 5 in early scabietic patients, whereas, it had negative correlation with IL - 1?, TNF - ?, and positive correlation with IL - 4 and IL - 5 in late ones. SOD showed negative correlation with IL - 1? and TNF - ?, and positive correlation appeared with IL - 4 and IL - 5 in both scabietic patients. Eosinophils (%) were positively correlated with all the studied cytokines in two groups of scabietic patients. The results also revealed that SDS - PAGE profile of the parasite heat stable proteins consisted of protein bands with molecular weights ranged from less than 10 to over than 100 kDa. Skin test demonstrated that (7/10, 70%) and (8/10, 80%) of scabietic patients had a positivity against 1.2 ?g/ml and 2.4 ?g/ml of sarcoptic mite extracts, respectively when prick tested. HDM extract was found to be positive in (4/10, 40%) of scabietic patients; while controls revealed negative result. It can be concluded that scabies affect TH1 and TH2 immune response as well as SOD activity and eosinophils. Sarcoptic proteins contain heat stable allergens which able to cause immediate type - 1 hypersensitivity when 1.2?g/ml of mite protein is skin prick tested, and there is a cross reactivity between Sarcoptes scabiei and HDM allergens

The study was conducted at the Faculty of Pharmacy - University of Kufa, and included using of 30 rats from Sprague - Dawley divided into five groups : the negative control group, the positive control group, the group treated with organic extract, the quercetin treated group and the group treated with extract and quercetin together. The period of dosing continued for 7 days and on the eighth day all groups(except the control group) were poisoned with carbon tetrachloride. After 24 hours the animals were killed. The study lasted from April 2013 - until July 2013. The Liver weights of treated animals were measured, the effectiveness of transferase enzymes Alanine Transaminase, Aspartate Transaminase and Alkaline phosphatase and total bilirubin level in the serum were assessed, also the Glutathione and Malondialdehyde levels were estimated in liver homogenized. The results showed the following : A significant increase (P<0.05) in the liver weights, the effectiveness of the amine transferase enzymes, Alkaline phosphatas and total bilirubin in the serum of the treated animals when compared with the negative control group, while these parameters revealed a significant decrease (P<0.05) at the treated groups as compared with the positive control group. The results showed no significant difference (P>0.05) in the Glutathione level and a significant increment (P<0.05) in the level of Malondialdehyde in the treated groups when compared with the negative control group. Also, the results showed a significant decrease and increase (P<0.05) in the levels of Glutathione and the Malondialdehyde at the treated animals as compared with the positive control group. The present study showed numerous histological changes which resulted from the injection with carbon tetrachloride. In relation with the tissue sections which have been taken from the treated groups : with the extract, quercetin and the group treated with both revealed a protective effect for organic extract seeds of Trigonella foenum - graecum and quercetin drug on the liver tissues which had been poisoned and the best effect in the protection appeared at the group which was treated with the extract and quercetin together. From this study we can conclude that the Trigonella foenum - graecum organic extract and quercetin may reveal a protective role for the liver against carbon tetrachloride poisoning by maintaining the effectiveness of liver enzymes Alanine Transaminase, Aspartate Transaminase and Alkaline phosphatase and bilirubin level in the serum as well as by increasing the Glutathione level and falling the Malondialdehyde level in liver homogenized. Also we conclude that using Trigonella foenum - graecum organic extract and quercetin together led to double protection of the liver and efficiently as a result of the positive impact which resulted from using them together

Abstract: هدفت الدراسة الحالية الى تقييم انتشار جينات البيتا لاكتاميز ? - lactamases ومنها جينات البيتا لاكتاميز الواسعة الطيف lactamases - Extended Spectrum ? وجينات اخرى مثل جينات المقاومة للكوينولونات Quinolones المرتبطة بالبلازميدات plasmid mediated quinolones | The study aimed to evaluate the prevalence of plasmid mediated ? - lactamases including Extended Spectrum ? - lactamases and non - lactamases and study the horizontal gene transfer. A total of 800 of urine samples were taken from patient suffering from urinary tract infections had been collected during a period from February, to September of 2012, from the hospitals of AL - Najaf province. All the samples were cultured on MacConkey agar. From those 300 samples gave positive bacterial growth , 250 were lactose fermentative isolates, which were submitted to conventional tests including IMVIC and motility tests ultimately lactose fermentative, non motile isolates were candidate to Vitek 2 system to confirm the identification. the results revealed that there were 42 (16.8%) of isolates identified as Klebsiellae represented by Klebsiella pnumoniae ssp pneumoniae, 9(3.6%) of isolates diagnosed as Raoultella ornithinolytica, and 1(0.4%) for both K. pnumoniae ssp.ozeanae and Raoultella planticola. Screening tests were performed , disk diffusion test revealed different pattern of resistance, screening of ESBLS by MacConkey agar medium supplemented with 2 mg /l of Ceftazidime showed that 31 (58.5%) of isolates were initially ESBLS producers.Phenotypes confirmatory tests were conducted to different classes of antimicrobial agents, Disk synergism test revealed that 20(37.74%) of isolates were ESBLS positive, while Disk combination test by Ceftazidime + clav and Cefepime + clav revealed that 28(52.83%) and 43(81.13%) were ESBLS producers respectively, disk replacement test pointed that positive isolates were 26(49%), vitek2 system revealed 33(62%) of isolates were ESBLS producers.Imipenem - Ceftazidime antagonism test revealed that there was no isolate produce induced AmpC beta - lactamase, AmpC disc test revealed that no one of isolates were AmpC producer. The result of MHT(Modified Hodge test) revealed that all isolates were not Carbapenemases producers. Molecular study of different antimicrobial resistance genes were performed, the results reveald high percent of occurence as follow : blaTEM genes (90.6%), blaSHV gene(81.13%), blaCTX - M genes (88.6%), sul genes (88.6%), aac(6')Ib - cr genes (84.9%) and qnr - B genes (41.5%).the study also highlighted an association between studied genes. Finally many attempts to study of gene transfer by conjugation were conducted but all of them were failed, except one isolate (No. 5) was succeeded with frequency of conjugation (0.5×10 - 7). This study concluded that there were high prevalence of some plasmid mediated genes of isolates with clear multi gene resistance patterns as compared with some other genes which propose the high selective pressure of these genes and isolates might acquired resistance by mobile elements such as plasmids and integrons.

Abstract: قدر التنوع الوراثي لـ 19 من الانماط الوراثية للطماطة (المحدودة وغير المحدودة النمو) المستزرعة في العراق باستخدام اثنين من واسمات الدنا (DNA Markers) المعتمدة على تفاعلات البلمرة المتسلسلة Polymerase Chain Reaction (PCR) وهما واسمات التفاعل التضاعفي العشوا | Genetic diversity of 19 tomato genotypes (determinate and indeterminate) cultivated in Iraq using two polymerase chain reaction based DNA markers (PCR based DNA markers); Random Amplified Polymorphic DNA (RAPDs) and Simple Sequence Repeats (SSRs).Variation of some growth criteria and morphological traits for each genotype were recorded in the growing season of 2012 - 2013.High variability was observed in plant height, leaf area, number of inflorescence, number of flowers and fruit weight among genotypes To achieve PCR reactions, total genomic DNA was isolated from fresh leaves (2 weeks old). The average yields of DNA were in the range of 100 - 295 ng/?l with a purity ranging between 1.8 - 1.9.RAPDs amplifications were performed for genotypes fingerprinting by testing 27 Operon primers. DNA polymorphisms among genotypes were scored within detectable amplified fragments (their numbers and molecular weight) after agarose gel electrophoresis and staining with ethidium bromide. The 27 primers produced 442 of main bands, out of which 312 were polymorphic bands (70.5%) and 70 were monomorphic (15.8%) across all tested genotypes. Each selected primer produced between 60 bands (OPA - 14) to 290 bands (OPD - 13). DNA amplification products ranged in their size from 250 bp (OPA - 01, OPU - 14, OPX - 15, OPX - 19, OPT - 08 ( to 2755 bp (OPX - 18). The highest number of polymorphic bands (21 bands) was produced by primer OPU - 03, while the lowest number of polymorphic bands (3 band) was produced by both primers OPA - 14 and OPB - 17.The primers varied in their capacity in producing polymorphic amplified profiles among tomato genotypes which individually reflected genotype specific DNA profiles (fingerprints). The most important primers for this purpose were primers that produced more variety specific DNA profiles, such as OPD - 13, OPT - 08, OPW - 04, OPA - 04, OPA - 15, OPB - 18, OPU - 03, OPC - 09.The highest value of discrimination among genotypes in this study was obtained by primer OPU - 03, while the lowest discrimination value was produced by both primers OPA - 14 and OPB - 17. The primer efficiency ranged from 0.13 in (primer OPC - 09) to 0.02 in (primer OPB - 17). The lowest genetic distance was 0.2294 between genotypes Oula and Shady lady, while the highest genetic distance was 0.9459 between genotypes Fotton and Special pack. Cluster analysis (Phylogenetic tree) by un weighted pair - group method of arithmetic means (UPGMA) based dendrogram revealed that they were two main genetic groups (major clusters). The first small major clusters included four (four genotypes) while the second large major cluster included (15 genotypes). A total of 21 alleles were detected among the tested genotypes using five SSRs loci distributed on four chromosomes of tomato. The molecular size of bands obtained from amplification of SSR products ranged from 121 to 247 bp. Alleles ranged from one in (Tom 8 - 9, Tom 41 - 42 and Tom 67 - 68 loci) to twelve in Tom 49 - 50 locus. The values of heterozygosity for each locus ranged between 0.63 for Tom 31 - 32 and 0.89 for Tom 49 - 50 with a mean value of 0.30. The polymorphic information content (PIC) values for the SSR loci ranged from 0.45 in Tom 31 - 32 to 0.58 in Tom 49 - 50 loci with an average of 0.21. Each one of (Tom 8 - 9, Tom 41 - 42 and Tom 67 - 68 loci) produce 0.0 value for both heterozygosity and PIC. The study revealed that, The lowest genetic distance was 0.3244 between varieties Tamara and W arda, while, the highest genetic distance was 0.9177between varieties Helam and Super marimond. The genetic similarity values ranging from 0.0823 to 0.6756 depending upon the genetic distance values that ranging from 0.3244 to 0.9177, indicating the largest diversity with percentage of 32 to 91% among the tested genotypes. The analysis of the results obtained from genetic distances and Neighbor - joining dendrogram (unrooted tree) revealed that, the 19 tested tomato genotypes can be grouped into two major groups : first cluster included nine varieties distributed in two subgroups. The second major cluster included 10 genotypes which in turn divided into two subgroups. The relationship among genotypes was not concern to their morphological characters and geographical origins. The overall analysis of the results show that both SSRs and RAPDs markers are powerful tools in fingerprinting and revealing the genetic relationships among tomato genotypes.

Abstract: هدفت الدراسة الحالية الى تقييم النسبة المئوية لكروماتين النطفة غير السوي Abnormal sperm chromatin percentage لمرضى عديمي الخصوبة مقارنة بالرجال الخصبين ودراسة العلاقة بين النسبة المئوية لكروماتين النطفة غير السوي ومعايير المني ومؤشر نوعية النطف وتركيز الم | The aim of the current study was to evaluate the Abnormal sperm chromatin percent for the infertile comparing with the fertile men and study the relation between the Abnormal sperm chromatin percent, the sperm and semen parameters, sperm quality index and MDA concentration for the infertile normozoospermic and asthenozoospermic patients comparing with the fertile men. This study was conducted in the laboratories of fertility centers in Al - Sader medical city/ Al - Najaf Al - Ashraf from 1 / 11 / 2012 to 30 / 4 / 2013.By the end of the semen liquefaction time, the 72 samples were divided into two groups : - the fertile men, (17), and the second is represented by the Infertile Normozoospermic (23) and the Asthenozoospermic (32). The age average was (1.36±30.9) , (0.98 ±29.6) and (1.63± 31.4) years respectively. The samples were restricted to the non smoking patients only. The study results show a significant (P< 0.05) decrease in the sperm concentration, the percent of the progressive forward movement sperm, the sperm viability percent and the semen volume, and a significant (P<0.05) increase in the round cell concentration, liquefaction time, Malondialdehyde concentration and the percent of the abnormal chromatin of the Normozoospermic and the Asthenozoospermic patients comparing with the fertile men. The results also show a negative correlation (P<0.05) between the (MDA) concentration and the parameters represented by the sperm concentration, percent of the progressive forward movement sperm, the normal sperm percent, the sperm viability percent sperm quality index and semen volume. While it had been noticed that there was a positive relation between the MDA concentration and the parameters represented by the sperm liquefaction time, round cell concentration and the abnormal sperm chromatin percent of all the study groups. In this research the relation between the sperm chromatin damage and the other study parameters had been studied by dividing the samples according to the degree of chromatin damage. The results show a significant (P<0.05) decrease in the sperm concentration, the percent of the progressive forward movement sperm, the normal sperm percent, the sperm viability percent, the sperm quality index and semen volume, with a significant (P<0.05) increase in the liquefaction time and round cell concentration (MDA) concentration and the percent of the abnormal chromatin of the group (> 10) comparing with the group (? 10) for the fertile men, also the results show a significant (P<0.05) decrease in the sperm concentration, the percent of the progressive forward movement sperm, the normal sperm percent, the sperm viability percent and the semen volume, with a significant (P<0.05) increase in the liquefaction time and round cell concentration for the group (> 25) comparing with the group (? 25) for the infertile Normozoospermic patients. For the Asthenozoospermic patients the study shows a significant (P<0.05) decrease in the sperm concentration, the percent of the progressive forward movement sperm, the normal sperm percent, the sperm quality index, the sperm viability percent and semen volume, and a significant (P<0.05) increase in the liquefaction time and round cell concentration for the study groups represented by the group (> 20 - 40) and (< 40) comparing with (? 20). From the above results it could be concluded that the increase of (MDA) level resulted from the active (ROS) leads to an increase in the abnormal chromatin percent of the infertile Normozoospermic patients which had a negative effect on the semen parameters which could be a reason for the infertility for those patients

Abstract: يعد السرطان واحدا من المخاطر الاساسية التي تهدد حياة الانسان في مختلف بلدان العالم، لان هذا المرض لا يقف عند عضو معين فهو ينتشر الى كثير من اعضاء الجسم الاخرى ليفتك بها، ومن هذا انطلقت الدراسة الحالية التي اجـريت للفترة من تشرين الاول / 2012 م ولغاية اب | The Cancer is one of highly risk factors that threats the human life world wide, because the disease has metastatic property from primary to secondary sites. This study was carried out during the period extending between October 2012 and August 2013, in laboratories of Faculty of Veterinary Medicine & Faculty of Education for girls in University of Kufa. It is aimed to follow up the changes in volume of growing tumor in female albino mice which were injected with cells of murine mammary adenocarcinoma. The histological changes, immunohistochemical examination investigate the distribution of CD8+ T lymphocytes were also studied. The changes of IgG and IgM titers in serum were also investigated. The study included use of 40 female albino mice at age 6 - 8 weeks, 25g weight average. All mice were injected by cells of murine mammary adenocarcinoman (AM3) in subcutaneous of femoro - dorsal region, and ascending to cervical region. The tumor was successfully implanted into 28 mice. These mice divided into four groups, each contains seven tumor - bearing mice. The animals of the first group subjected to the investigations after 48 hrs from implantation , while those of 2nd, 3rd & 4th groups after 10, 20, 30 days respectively. Seven healthy mice were considered as control group to comparison of immunological tests. Tumor volume & relative tumor volume were measured in different experimental groups. Histological section were prepared by routine histological techniques that including the steps of dehydration, clearing, infiltration, embedding, cutting & staining by hematoxylin and eosin stains, then microscopical examination had been done by optic microscop. The results indicated there was significant increase in tumor volume and relative volume with advancing of tumor age in animals of 2nd, 3rd & 4th groups where compared with first group. The gross examination of the animals of different groups showed that the tumor grew as irregular projection with loss of hair and erosion of overlying skin with milky discharge. The post - mortem examination revealed that the tumor mass was divided into small lobules with various coloures and the tumor mass surrounded by fibrous tissue which increase in thickness as the tumor increased in the age. Extensive areas of necrosis were noticed in the centers of the tumor masses. The microscopic examination of tumor masses in the first group raveled there were sub cataneous small aggregation of tumor cells at the implantation region as well as dispersion of the implantated tumor cells. The inflammatory cells, especially neutrophils were infiltrated. In the second group, the tumor masses showed lobulation by fibrous septa with proliferation of new blood vessels with congestion of them. Infiltaration of inflammatory cells, particularly the lymphocytes. The tumor cells arranged as ribbons and acini, characterized by hyperchromacia pleomorphism, high N/C ratio as well as appearance of mitotic figures. The histological features of the third group revealed increase of amount of fibrous tissue extensive necrotic areas and loss of architecture. These appeared in the fourth group in addition to the presence of cellular debris with dead inflammatory cells. The immunohistochemical study showed no CD8+ T cells were infiltrated in the mass of first group but the little infiltration of these cells was show in the second group. The highest infiltration of the CD8+ T cells was appeared in the third group. while the little cells were discharged in the fourth group. The results of the single radial immunodiffussion assay showed that the implantation with mammary adenocarcinoma had significant effect in increasing the titer of IgM after (48) hours and (30) days from implantation of tumor compared to control group, while the titer is not significant after (10) and (20) days from implantation of the tumor compared to control group. Also the results revealed a significant decrease in the titer of IgG after (48) hours and (30) days from implantation of the tumor, while were no significant increase in the titer of immunoglobulin after (10) and (20) days from implantation of the tumor, compared to the control group. From all the above, We concluded that there was variable histological changes as the age of tumor advanced these changes were characterized by increase of amount of tumor cells in the first groups and occurrence of necrosis in late groups, as well as increase the fibrous tissue as the tumor age advanced. The infiltrative CD+8 lymphocyt was also variable as change of tumor age. The implantation of tumor in mice groups had marked effect in stimulation of humoral immune response through increase of IgM & IgG titers.

Studies concerning the effect of Iraqi propolis are rare. Therefore, the goal of this work is to analyse the effect of Iraqi propolis on some hematological and biochemical parameters in alloxan - induced diabetic rabbits, as well as study the histological observations of the pancreas, liver and kidney. Female local rabbits (Oryctolagus cuniculus) were used for this study. The overall number of animals used was 30.They were randomly divided into five groups. Diabetes was induced in all rabbits, except normal control, by a single dose of alloxan (150 mg/kg, i.v.). Development of induced diabetes mellitus was confirmed on first week after alloxan administration by examining the fasting glucose level in the blood taken from marginal ear vein. Rabbits with glycaemia were treated with alcoholic extract of propolis for 23 days. Diabetic control group did not treat with propolis. The treated animals were subdivided into three groups according to the dose of propolis extract. Three oral concentrations of propolis extract were investigated (50, 100 and 200 mg/kg/day).The following parameters have been studied were changes in weights of body, liver and kidneys ; changes in the hematological values that include erythrocytes, hemoglobin, leukocytes and hematocrit also changes in the biochemical values which included glucose, total protein, triglycerides, total cholesterol, aspartate transaminase (AST), alanine transaminase (ALT), blood urea, creatinine and Malondialdehyde (MDA), in addition histopathological study of pancreas, liver, and kidneys. The results indicate a significant decrease (P < 0.05) in the body weight of alloxan - induced diabetic rabbits in comparison with control group, while there were significant increases in the weights of liver and kidneys. The present study showed that alloxan induced significant decreases (P<0.05) in all primary blood indices; erythrocytes, hemoglobin, packed cell volume (hematocrit) and leukocytes. Also, serum biochemical changes showed significant increases (P<0.05) in glucose, total protein, triglycerides, total cholesterol, AST, ALT, blood urea, creatinine, and MDA comparison with control group.Histopathological changes in pancreas, liver and kidneys, observed microscopically, revealed degrees of damage in the tissues, while these organs of control groups exhibited a normal architecture. The treatment with alloxan resulted in several forms of histological alterations such as cytoplasmic vacuolation, degeneration, necrosis, cell hypertrophy, nuclear diploid, diffusion of inflammatory cells, dilatation in ducts, stagnation in secretory fluid. in addition, hemorrhage, dilatation and congestion in blood vessels and disorganization of histologic architecture, Generally, the gradual improvement in blood values was noticed with the increase in concentration alcoholic extract of propolis and return back the normal histological shape of pancreas, liver and kidneys. Propolis extract in rabbits had a potent antihyperglycemic effect, antioxidant activities, radical - scavenging capacities, tissue regeneration properties, and that may be due to the high biological activity and nutritive values contents in bee propolis. In conclusion, the results suggest that propolis could potentially contribute for the prevention and treatment of diabetes mellitus.

وقد ظلت اسباب نشوء هذه الحالة غير واضحة المعالم, اذ اعزيت بعض من اسبابها الى ا | Preeclampsia is considered as one of the serious complication in the third trimester of pregnancy. It is associated with hypertension and proteinuria. The causes of preeclamptic are still not well understood. Evidences suggested the involvement of metabolic related mutations in the disease. In the current study, the genetic mutations of CBS and MTHFR genes are studied as markers for the diagnosis of preeclampsia. Sixty preeclamptic pregnant women and forty healthy pregnant women as control group diagnosed by the physicians at Azzahra Teaching hospital for obstetrics &pediatrics in Najaf province from December 2012 to August 2013 The poly merase chain reactions technique was used to evaluate the incidence and frequency of C?S &MTHFR mutations. A questionnaire was conducted to obtain the information of the patients and the control group. Genetic mutation were detected by polymerase chain reaction (PCR)and agaros gel electrophoresis. The information indicated that the age of pregnant women, gestation age, previous history of the patients, family history and parity are involved in the pathogenesis of preeclampsia. The results of the present study of preeclampsia showed a significant increase (P ? 0.01) in patients (25 - 34) years old when compared with the other group. where the results found a significant increase (P <0.01) of Preeclampsia occurred during (33 - 39) weeks of pregnancy. And in the patients with previous history for preeclampsia there was significant increase (P<0.001) compared with healthy pregnants. Pregnants with preeclampsia in the family history showed significant increase (P< 0.01) as for women pregnant for the first time showed and significant (P<0.01) and constituted 8% pregnanst with preeclampsia who had diabetes mellitus. The extraction and quantification of cell free fetal DNA (cff DNA) revealed a significant (P<0.0001)increase in preeclamptic patients when compared with those of healthy pregnant women. It is confirmed by the use of an internal negative control through the monitoring of SRY gene (Y chromosome gene) amplification that the gene belong to the infant and 40 pregnant woman were followed up till the labor. Thus, 9 of them had delivered males, while 30 had delivered females. The wild type SRY gene band was obtained clearly from those of male delivery, but not from those of female delivery. Mutations of C?S gene were analyzed by theuse a primer intended to amplify a fragment of171 bp.The rate of mutation was found to be 51.7% and 5% in preeclamptic patients and healthy pregnant women respectively and Odd Ratio=20.31, P value=10 - 7. The C677T mutation of MTHFR gene was verified by the use of a primer designed to amplify a fragment of 198bp. The prevalence of mutation was indicated to be 68.3% and 17.5% in the preeclamptic patients and healthy pregnant women respectively and Odd Ratio =10.17, Pvalue =6.2*10 - 7. Results demonstrated the wide involvement of C?S and MTHFR gene mutation in the pathogenesis of preeclampsia, but they are no crucial for the development of the disease. In addition there are so many factors thatmay be involved in such development.
