Abstract: Summary ►Urinary tract infections (UTIs) are the commonest human bacterial infections and are responsible for substantial morbidity and mortality, resulting in increased healthcare costs. Specialized Escherichia coli strains referred to as Uropathogenic E. coli (UPEC) causes most UTIs. ►Total 120 isolates primarily identified as E.coli; were collected from (December 2018 to February 2019). Out of total collected isolates, 94/120 were included in this study and the contaminated bacteria or weak growth were excluded. The isolated pure colonies were re-identified using some standard morphological tests followed by genetically identification using housekeeping gene (HKG) 16S rDNA. The results confirm that all the isolates were definite as E. coli. ►The UPEC isolates showed their ability of adherence to uroepithelial cells. The aggregation ability of bacterial cells were appeared as dotes on the surface of epithelial cells. ►The antimicrobial susceptibility test (AST) was performed for 50 isolates under study. The highest resistance rate was concerning to Aztreonam; Cefazolin; Ceftazidime and Ceftriaxone (84%), Cefepime (82%), while the minimum resistant was for Imipenem; Ertapenem; Meropenem; Amikacin when it was (2%) beside that none of the isolates was able to resist Tigecycline. According to AST for fifty UPEC isolates; (88%) were multidrug resistance (MDR), while (2%) was extensively drug resistance (XDR) and (10%) were sensitive to all antibiotics thus they were non-drug resistant (NDR). ►All isolates were subjected to detect type1 fimbriae genes (fimA operon) by polymerase chain reaction using specific primers (designed in this study) followed by sequencing the amplified fragment and analyzed using Geneious prime software. The results showed that all the isolates (100%) were positive for fimA, fimI, fimC, fimD, fimG, fimH, while (96%) for fimB, fimF and (82%) for fimE. ►Raw milk samples were collected for probiotics bacteria isolation. All isolates were subjected to the cultural, microscopically, biochemical tests and Vitek2 system. Results indicated that six isolates of Lactococcus spp. Identified as: Lactococcus lactis ssp. lactis, in addition, 14 isolates of Leuconostoc spp. distributed among species as (8) isolates belonged to Leuconostoc mesenteroides ssp. cremoris (3) isolates belonged to Leuconostoc mesenteroides ssp. mesenteroides, (2) isolates identified as Leuconostoc pseudomesenteroides and only one as Leuconostoc citreum. ►All isolated probiotics bacteria were tested for Titanium dioxide (TiO2) nanoparticles production. The results showed that they had the ability to biosynthesize TiO2 NPs, biosynthesized TiO2 NPs were characterized using UVVis Spectroscopy; Atomic Force Microscope (AFM); X-ray Diffraction (XRD); Field Emission Scanning Electron Microscopy (FE-SEM) Energy dispersive Xray spectroscopy (EDX); and Fourier Transform-Infrared Spectroscopy (FTIR). The detected absorption spectrum with a wide-ranging absorption band in UV range and its cut off wavelength was (344-348) nm for all 20 probiotic isolates. ►The smoothness and roughness of all samples of biosynthesized TiO2 NPs was detected by AFM with smoothest sample for L. mesenteroides ssp. cremoris (Leu.4) as 1.137nm. XRD results indicated the formation of nano anatase crystals. ►The Scanning electron microscope images of biosynthesized TiO2 NPs biosynthesized by probiotics bacteria; that were consists of a uniform distribution of spherical shaped nanostructure crystals with a smallest size of (37.48) nm for L. citreum (Leu. 13). ►The EDX analysis of TiO2 NPs existing in medium which confirm that all the samples contained the elements (Ti, O), which again established that the biosynthesized NPs were pure TiO2. ►The determination of functional group of biosynthesized TiO2 NPs studied using FTIR. The main IR absorption peaks of TiO2 biosynthesized by probiotics bacteria were in range at (408- 445) cm-1; these peaks belonged to anatase Titania. ►The biosynthesized TiO2 NPs at concentrations (1000- 0.001) mg/mL were tested for its antibacterial activity against UPEC isolates, thus there was neither bactericidal effect against pathogenic bacteria nor been as minimum inhibitory concentration (MIC). ►There was no effect of temperature, pH and photocatalytic activation on antibacterial activity of biosynthesized TiO2 NPs against clinical UPEC isolates. ►Regardless of no growth inhibition activity showed on culture media after subjecting bacterial UPEC isolates to TiO2 NPs. There was a noticeable inhibitory effect of TiO2 NPs on the attachment ability of bacterial cells without killing them. The highest inhibition was recorded at the higher concentration of biosynthesized TiO2 NPs (1000 g/mL) with inhibition reached 92% and 90.3%, while the lowest inhibition were 52% and 54.8% at 250g/mL against E.coli74 and E.coli23, respectively. ►The effect of biosynthesized TiO2 NPs by probiotics bacteria and commercial TiO2 NPs was studied on the expression of fim genes. There was a subsequent change in the gene expression in term of folding and CT values when the expression of fimH, C, D genes start to decrease by increasing concentration of TiO2 NPs as compared with control and the maximum inhibitions was noticed by using 1000 g/mL concentration of the NPs specially against fimD.

Abstract: يعتبر مجمتع الطيطوي مؤشرات حيوية وهي انواع توفر معلومات تتعلق بالجودة البيئية وتستخدم لتقييم جودة الاراضي الزراعية، وهي حساسة للتغيرات في استخدام الاراضي وتدهور الموائل، وقد يشير وجودها او غيابها الى صحة النظام البيئي. هناك معلومات نادرة عن مجتمع الطيطوي وحياتية تكاثره في العراق. اجري مسح حقلي من 2022 سبتمبر الى اغسطس2023 ، باجمالي 72 مسح حقلي في منطقة المالح في شمال بابل – وسط العراق، والذي يعتبر من الاراضي الرطبة ، وتبلغ مساحته حوالي 2000 هكتار (20 كم2). هدفت الدراسة الى الحصول على دراسة بيئية وحياتية لمجتمع طيور الطيطوي (Vanellus spp.) في منطقه المالح في محافظة بابل. ولاجل تحقيق عدة اهداف تضمنت: تصنيف انواع الطيور في منطقة المالح و تحديد مجتمع الطيطوي مع حجم السكان، دراسة التحليل الكمي لتماثل بيوض مجتمع طيور الطيطوي والاختلافات بين مجموعات انواع الطيطوي ودراسة العوامل البيئية (الاس الهيدروجيني ورطوبة التربة وحرارتها وقوامها) لاعشاش طيور الطيطوي استحدمت طرق المسح الحقلي اخذ العينات عن بعد والمعتمدة على احتمال اكتشاف طيور الطيطوي واجريت طرق المسح الميداني هذه من اجل الحصول على تقديرات موثوقة لكثافة الانواع وحجم السكان طيور الطيطوي في منطقة المالح, تم تحديد اجمالي اربعة خطوط عرضية تغطي منطقة دراسة تبلغ مساحتها 1100 هكتار (11 كم2). سجلت 122 نوعا في المسوحات الميدانية الحالية في منطقة المالح تنتمي الى 17 رتبة و38 فصيلة، و ان 7 انواع ضمن القائمة الحمراء للاتحاد الدولي لحفظ الطبيعة وتمثل انواع الطيور حوالي 30.65% من العدد الاجمالي للطيور العراقية وتم تسجيل 4 انواع من جنس الطيطوي (Vanellus spp.). سجل 904 فردا من اربعة انواع من طيور الزقزاق (Vanellus spp.) ينتمون الى رتبة Charadriidae، عائلة Charadriiformes بين 5 انواع موجودة في العراق.(الزقزاق الشمالي V. vanellus,78; ناتىء الجناح V. spinosus,531; احمر اللغد V. indicus,136; and ابيض الذيل V. leucurus,159). استضافت منطقة المالح مجتمع طيور الطيطوي بحجم سكان المكون من اربعة انواع، الزقزاق الشمالي V. vanellus (148.59-203.45) في شهري كانون الاول و تشرين الاول 2022 :ناتىء الجناح V. spinosus (270.74-1248.73) في نيسان و اب 2023; احمر اللغدV. Indicus (277.17-931.67) في تشرين الاول 2023 و كانون الثاني 2023; ابيض الذيل V. leucurus (163.64-742.69) في تشرين الاول 2022 ويوليو 2023

Abstract: الملخص | تضمنت هذه الدراسة تقييم اثار اضافة مبيد الكلوربيريفوس ومستويات مختلفة من مسحوق اوراق المورينجا اوليفيرا Moringa oleifera في اسماك الكارب الشائع Cyprinus Carpio ودراسة تاثير مسحوق اوراق المورينجا اوليفيرا في تقليل سمية الكلوربيريفوس، اجريت التجربة في مختبرات كلية الزراعة/ جامعة الانبار، استمرت التجربة لمدة 60 يوما من 25 / 8 / 2022 لغاية 23 / 10 / 2022. بضمنها مدة الاقلمة. | وزعت 144 سمكة بمعدل وزن 49.29 ± 0.44 غم عشوائيا الى ثمان مجموعات تحتوي كل منها على 18 سمكة، كل مجموعة تمثل معاملة، كل معاملة وزعت على ثلاثة احواض زجاجية بواقع 6 اسماك للحوض الواحد كمكررات للمعاملة. اجريت تجربة تحديد قيمة التركيز النصفي القاتل LC50، وقد بينت النتائج بعد 96 ساعة ان قيمة LC50 هي 0.60 ملغم\لتر وخفف المبيد بنسبة 1/10 ليصبح تركيزه 0.06 ملغم\لتر. غذيت الاسماك بمستويات مختلفة من اوراق نبات المورينجا اوليفيرا. تم اعتبار المجموعة الاولى T1 كمجموعة مقارنة بدون اضافة مسحوق اوراق المورينجا ، وعرضت اسماك المعاملة الثانية T2 لمبيد الكلوربيريفوس بدون اضافة مسحوق الاوراق كمجموعة مقارنة موجبة، في الوقت الذي اضيف فيه مسحوق الاوراق الى المجاميع T3 وT4 وT5 بكميات 5 و10 و15 غم\كغم على التوالي بدون تعريضها للمبيد. كما اضيفت النسب نفسها من الاوراق للمجاميع T6 و T7 و T8، مع التعرض للتركيز نفسه من المبيد، غذيت الاسماك مرتين يوميا بنسبة تغذية 2% من وزن السمكة. | اظهرت نتائج دراستنا الحالية ان اعلى معدل نمو كلي ومعدل نمو يومي ومعدل نمو نسبي حدث في المعاملة الرابعة T4 والتي سجلت 49.49 و0.87 غم و102.51 % على التوالي. كما سجلت المعاملة الثالثة T3 اعلى معدل نمو نوعي 1.23 %\يوم. في حين ان اكبر انخفاض في المعدلات ذاتها حدث في المعاملة الثانية T2 التي سجلت -4.64 و-0.08 غم و-8.66 % و-0.16 %\يوم على التوالي. بينما كانت افضل معالجة في المعاملة السابعة T7 التي سجلت نمو كلي ويومي ونسبي ونوعي بلغت 41.07 و0.72 غم و84.84 % و1.07 %\يوم على التوالي. | سجلت كريات الدم البيض اعلى معدل لعدد الكريات في المعاملة الثانية T2 المعرضة للمبيد اذ سجلت 7.90 × 310 ، في حين ان اقل معدل لها كان في المعاملة الرابعة T4 التي سجلت 4.94 × 310. بينما سجلت المعاملة السابعة T7 افضل معالجة، فقد سجلت 5.35 × 310. فيما يتعلق بمؤشرات كريات الدم الحمر فقد سجلت المعاملة الرابعة T4 اعلى معدل لعدد الكريات بلغ 3.34 × 610 واعلى تركيز للهيموغلوبين بلغ 12.67 غم\100 مل، واعلى نسبة لحجم الكريات المضغوطة بلغت 35.04 %، بينما كانت اقل معدلات كريات الدم الحمر وهيموغلوبين وحجم دم مضغوط في المعاملة الثانية T2 التي سجلت 2.31 × 310 و 7.23 غم\100 مل و 19.67% على التوالي. كانت افضل معالجة لكل من معدلات كريات الدم الحمر وهيموغلوبين وحجم دم مضغوط في المعاملة السابعة T7 التي سجلت 2.77 × 610 و 10.29 غم\100 مل و29.84 غم\100 مل على التوالي. | بينت نتائج الاختبارات الكيموحيوية ان المعاملة الثانية T2 سجلت اعلى المعدلات لفعالية انزيمات الفوسفاتيز القاعدي ونازعة الالانين ونازعة الاسبارتيت واختبارات وظائف الكلى اليوريا وحامض اليوريك والكرياتينين والمالونالديهايد فضلا عن معدلات الكوليسترول والبروتينات الدهنية منخفضة الكثافة والدهون الثلاثية ، اذ سجلت فعاللية انزيم الفوسفاتيز القاعدي معدلا بلغ357.89 وحدة دولية\لتر، ونازعة الاسبارتبت معدلا بلغ 284.22 وحدة دولية\لتر ، ونازعة الالانين معدلا بلغ 63.33 وحدة دولية\لتر ، يوريا بلغت 19.78 ملغم\100 مل ، حامض اليوريك بلغ 2.01 ملغم\100 مل ، كرياتينين بلغ 0.39 غم\100 مل ، والمالونالديهايد بلغ0.61 نانومول\مل ، كولسترول بلغ 214.00 ملغم\100 مل ، والبروتينات الدهنية منخفضة الكثافة بلغ 88.58 ملغم\100 مل ، والدهون الثلاثية بلغت 377.67 ملغم\100 مل. في حين سجلت المعاملة الرابعة T4 اعلى بروتينات دهنية عالية الكثافة بلغت 83.00 ملغم\100 مل. وسجلت المعاملة الرابعة T4 ايضا ادنى معدلات فوسفاتيز قاعدي بلغت 197.22 وحدة دولية\لتر، ونازعة الالانين بمعدل 36.78 وحدة دولية\لتر، وحامض اليوريك بمعدل 1.32 ملغم\100 مل ، والكرياتينين بمعدل 0.22 غم\100 مل. وقد سجلت المعاملة الخامسة T5 ادنى فعالية نازعة الاسبارتيت بمعدل 150.88 وحدة دولية\لتر، ويوريا بمعدل 9.67 ملغم\100 مل ، والمالونالديهليد بمعدل 0.33 نانومول\مل، وكوليسترول بمعدل 150.67 ملغم\100 مل، والبروتينات الدهنية منخفضة الكثافة بمعدل 53.13 ملغم\100 مل. سجلت معاملات المعالجة بالمورينجا تحسنا في معدلات المؤشرات المدروسة، فقد سجلت المعاملة السابعة T7 معدل فوسفاتيز قاعدي بلغ 204.78 وحدة دولية\لتر ويوريا بمعدل 10.44 ملغم\100 مل وحامض يوريك بمعدل 1.39 ملغم\100 مل ، وكوليسترول بمعدل 156.33 ملغم\100 مل ، والبروتينات الدهنية عالية الكثافة بمعدل 64.33 ملغم\100 مل ، والبروتينات الدهنية منخفضة الكثافة بمعدل 57.78 ملغم\100 مل ، والدهون الثلاثية بمعدل 171.67 ملغم\100 مل. | اظهرت الفحوص النسجية حدوث ضرر في انسجة الغلاصم والكبد والامعاء الدقيقة والكلي في الاسماك المعرضة للمبيد. كما تبين ان النسب المنخفضة والمتوسطة من مسحوق اوراق المورينجا ( 0.5 % و 1 % ) كانت ذات تاثيرات ايجابية، مع وبدون التعرض للمبيد، في حين سببت اضافة لنسب مرتفعة من مسحوق الاوراق (1.5 % ) تضرر الانسجة المدروسة، وزادت نسبة الضرر مع التعرض للمبيد. | يستنتج من هذه الدراسة ان مبيد الكلوربيريفوس شديد السمية بالنسبة لاسماك الكارب الشائع، وان اضافة نسب معتدلة من مسحوق اوراق المورينجا اوليفيرا له دور في تحسين الحالة الصحية للاسماك عن طريق تعزيز معدلات النمو والمعايير الدمية والفسيولوجية والكيموحيوية والصفات النسيجية لاسماك التجربة. |
