دراسة نسيجية وكيموحيوية لبعض الديدان الشريطية في مضائف فقرية مختلفة == Histological and Biochemical Studies for some Cestode Tapworm in different Hosts

Abstract: تضمنت الدراسة الحالية البحث عن الديدان الشريطية البالغة في اربعة انواع من المضائف الفقرية بعد تشريحها باعداد بلغت ( 235 ) سمكة من عائلة الشبوطيات Cyprinidae ومن نوع الحمري Barbus luteus و( 15 ) افعى من عائلة Hydrophiidia ونوع Microcephalophis gracilis و( 25 ) من طائر النورس من عائلة Loridae ونوع Laurus canus وكان عدد الاغنام ( 95 ) من عائلة Ruminant ونوع Ovis ovis .تم الكشف عن الجهاز العصبي لاربعة انواع من الديدان الشريطية باستخدام الطريقة الانزيمية وبتقنية ( AchI ) اذ بينت النتائج ان هناك اختلافا في الاجهزة العصبية المركزية Central nervous system (CNS) والاجهزة العصبية المحيطية Peripheral nervous system وفيما يلي وصف لجهاز العصبي لكل نوع من هذه الديدان : Bothriocephalus sp. : يتكون الجهاز العصبي المركزي من زوجين من العقد الدماغية احداهما كبيرة تقع في منطقة التعقر الممصي ترتبط مع بعضها بحلقة دماغية Cerebral ring والاخرى صغيرة تقع في مقدمة الرؤيس وعند القرص القمي وترتبط مع بعضها بحلقة قمية Apical ring ، وترتبط كل من الحلقتين الدماغية والقمية برابط عصبي حلقي Ring nerve commissure وتنشا من العقد الدماغية الكبيرة والعقد الصغيرة عدد من الاعصاب لتزود التقعر الممصي ومقدمة الرؤيس . اما الجهاز العصبي المحيطي فيتكون من جذع عصبي رئيس ينشا من العقد الدماغية الكبيرة ثم يتفرع الى زوج من الجذوع الجانبية السميكة في كل جانب من جسم الدودة ، كما وينشا من العقد الدماغية القمية خمسة ازواج من الحبال العصبية الجانبية Lateral nerve cords تقع الى الخارج من الجذوع العصبية السميكة وعشرون عصبا وسطيا ينشا من العقد الدماغية الكبيرة . ترتبط الجذوع الجانبية السميكة والحبال العصبية الجانبية والاعصاب الوسطية بعدد من الروابط المستعرضة . كما تميزت هذه الدودة بظهور شبكة عصبية في كل قطعة وامتدادها الى الحافات الخارجية لجسم الدودة .Ophiotaenia europaea : من زوج من العقد الدماغية Cerebral ganglia تقع في منطقة المحاجم والتي ترتبط مع زوج اخر من العقد القمية اصغر منها تقع في منطقة العضو القمي ، وترتبط العقد القمية مع العقد الدماغية بوساطة رابط حلقي . وينشا من كل عقدة عدد من الاعصاب الصغيرة والنحيفة لتعصيب حافة الرؤيس والمحاجم والعضو القمي .ينشا من العقد العصبية الدماغية زوجين من الجذوع العصبية الجانبية السميكة ، كما وينشا من العقد القمية اربعة ازواج من الحبال العصبية الجانبية وقد لوحظ عدد من الروابط العصبية تتراوح ما بين ( 6 - 8 ) في كل قطعة جسمية كما لوحظ وجود رابط عصبي سميك يفصل كل قطعة جسمية عن الاخرى .انفردت الدودة الحالية بتوزيع العقد العصبية الجانبية الخلفية على الجذوع العصبية الجانبية والحبال والاعصاب الوسطية .Raillietina echinobothrida : يتكون الجهاز العصبي من عقدتين دماغيتين كبيرتين Cerebral ganglia ترتبطان برابط عصبي دائري وتنشا منهما ( 4 - 6 ) من الاعصاب الامامية الصغيرة لتزود المحاجم وعضلات الخطم وينشا من هذه العقد زوجين من الجذوع العصبية الجانبية السميكة جدا Thick lateral nerve trunck وكذلك اربعة ازواج من الحبال العصبية الوسطية Medial nerve cords . ترتبط الجذوع العصبية السميكة والحبال العصبية الوسطية بعدد من الروابط المستعرضة تتراوح بين ( 6 - 8 ) من الروابط العصبية السلمية وقد شوهد رابط عصبي مستعرض سميك يفصل القطع الجسمية بعضها عن البعض الاخر . كما شوهد اضمحلال العصب في منطقة القطع الحبلى .Moniezia expansa : يتكون ال جهاز العصبي المركزي من زوج من العقد الدماغية كثيرة الاجزاء تدعى بمعقد الدماغ ترتبط مع بعضها بحلقة دماغية Cerebral ring يخرج منها عددا من الاعصاب الامامية النحيفة لتزود المحاجم والخطم والحافات الخارجية للرؤيس مكونة شبكة عصبية للرؤيس . كما يخرج من هذه العقد العصبية زوج من الجذوع الجانبية السميكة وزوجين من الحبال العصبية والتي تتميز بسمك اقل من الجذوع الجانبية وقد شوهد ان الحبال العصبية الوسطية تمتد لعدة قطع جسمية ثم تضمحل عند بدء القطع غير الناضجة لتصبح بشكل اعصاب نحيفة وسطية وتتشابك مع الروابط مكونة الشبكة العصبية والتي تنتهي بعقيدات عصبية . كما شوهدت غدد ما بين القطع الجسمية Interproglottid glands واقعة على الرابط المستعرض السميك الذي يفصل القطع الجسمية بعضها عن البعض الاخر وان اعلى فعالية لانزيم الاستيل كولين استريز ( AchE ) توجد في الغدد ما بين القطع .اظهر الفحص بالمجهر الالكتروني النافذ لانسجة الديدان الشريطية قيد الدراسة بان الجليد فيها يتكون من طبقتين رئيستين خارجية وداخلية تفصل بينهما طبقة العضلات ويغطى الجليد ببروزات سايتوبلازمية تدعى بالشعيرات الدقيقة Microtriches تبين انها على ثلاثة انواع خيطية ومخروطية واصبعية وقد ظهرت الاشكال المخروطية والاصبعية بصورة واضحة في الدودة الشريطية المسببة لخمج الاغنام Moniezia expansa كما ظهر الغشاء القاعدي Basement membrane بصورة واضحة .كما تطرقت الدراسة الى عدد من المتغيرات الكيموحيوية منها قياس تركيز البروتين الكلية ، الاحماض النووية ( DNA ، RNA ) ، الكاربوهيدرات ، الدهون ، الكوليسترول ، وفعالية انزيم الكلوكوز - 6 فوسفيت ديهادروجينيز ( Glucos - 6 - phosphate dehydrogenase (G - 6 - PDH) ) ، وانزيم الفوسفاتيز القاعدي (Alkaline phosphatase (AIP) ) والحامضي (Acid phosphatase, (AcP) ) وانزيم الكلوتاميت ترانس امينيز (Glutamat pyruvate transaminase (GPT) ) وكلوتاميت اوكزالواستيت ترانس امينيز (Glutamate oxaloacetate transaminase (GOT) ) فضلا عن انزيم الاستيل كولين استريز (Acetylcholinesterase (AchE) ) .اظهرت النتائج وجود اختلافات معنوية في تركيز البروتين ، DNA ، RNA ، الكاربوهيدات ، الدهون الكلية ، الكولسترول والانزيمات في هذه الدراسة وخاصة في الديدان الشريطية المسببة لخمج الطيور . وتبين ان هناك ارتفاعا في تركيز البروتين الكلي في الدودة الشريطية Moniezia expansa المسببة لخمج اللبائن اذ بلغ ( 248.667  3.2146 ) مايكروغرام / سم3 ، وانخفض الى ادنى مستوى في شريطية الطيورRaillietina echinbothrida اذ بلغ ( 97.000  1.000 ) مايكروغرام / سم3 في حين كان تركيز البروتين ( 151.667  2.0817 ) مايكروغرام/سم3 و( 195.000  13.2288 ) مايكروغرام/سم3 في دودة Ophiotaenia europaea وBothriocephalus sp. ، على التوالي ، في كل من الزواحف والاسماك ، وكذلك اظهرت النتائج ان تركيز الـ DNA كان اقل قيمة في دودة Raillietina echinbothrida ( 11.000  1.000 ) مايكروغرام/سم3 وارتفعت في الدودة Ophiotaenia europaea الى ( 12.333  0.5774 ) مايكروغرام/سم3 بينما كان تركيز الـ DNA ( 14.3667  0.5508 ) و( 24.333  2.0817 ) مايكروغرام/سم3 في دودة Bothriocephalus sp. وMoniezia expansa . اما تركيز الـ RNA في الدودة الشريطية Moniezia expansa فكان اعلى من بقية الشريطيات اذ بلغت ( 88.6667  3.214 ) مايكروغرام/ سم3 بينما بلغت ( 67.7133  1.353 ) و( 47.333  3.0551 ) و( 23.333  .2.0817 ) مايكروغرام/ سم3 في دودة Bothriocephalus sp. ، Ophiotaenia europaea وRaillietina echinbothrida على التوالي .في حين كانت كمية الكاربوهيدرات في الديدان المسببة لخمج الزواحف قليلة اما قيمتها في الديدان المسببة لخمج الاغنام فكانت اعلى مما هي عليه في بقية الطفيليات اذ بلغت ( 204.5796  2.0801 ) مايكروغرام/ سم3 ، ( 186.6000  9.8989 ) مايكروغرام/ سم3 ، ( 291.1267  1.9688 ) مايكروغرام/ سم3 ، ( 328.000  8.7178 ) مايكروغرام/ سم3 على التوالي في الديدان المسببة لخمج الاسماك ، الافاعي ، الطيور ، الاغنام .اما قيمة الكوليسترول فكانت قليلة في الديدان المسببة لخمج الطيور والافاعي اذ بلغت ( 32.1967  1.3550 ) ملغم/ديسيليتر ، ( 34.6833  2.2548 ) ملغم/ديسيليتر على التوالي . وقد تباينت قيم الدهون الكلية بصورة معنوية وكانت اعلى نسبة في دودة Moniezia expansa اذ بلغت ( 229.427  0.6839 ) ملغم/ ديسيلتر و( 169.0590  1.2694 ) ، ( 146.2080  0.7736 ) و( 114.4253  0.551 ) ملغم/ ديسيلتر في كل من Botheriocephalus sp. ، Ophiotaenia europaea وRaillietinia echinobothrida على التوالي .وتبين نتائج دراسة فعالية الانزيمات : (G - 6 - PDH), (AcP), (AIP), (GOT), (GPT), (AchE) .ان الدودة الشريطية Moniezia expansa تمتلك فعالية عالية للانزيمات ( G - 6 - PDH ) ، (AcP) ، (GOT) ، في حين كانت فعالية الـ (AIP) في الـ Bothriocephalus sp. اعلى من بقية الديدان وتلتها في الفعالية Moniezia expansa بينما في بقية الديدان كانت الفعالية منخفضة في المقارنة مع Bothriocephalus sp. والـ Moniezia expansa ، بينما كانت فعالية انزيم GPT متقاربة نوعا ما في جميع الديدان المدروسة ، اما فعالية انزيم Acetylcholinesterase ( AchE) فقد اظهر اعلى فعالية في الدودة الشريطية Raillietinia echinobothrida وتليها في الفعالية Moniezia expansa بينما كانت الفعالية في الـ Ophiotaenia europaea والـ Bothriocephalus sp. منخفضة مقارنة مع النوعين السابقين .وبينت دراسة الترحيل الكهربائي وجود حزم متباينة في العدد والسمك اذ بلغت اربع حزم في الدودة الشريطية المسببة لخمج الاسماك Bothriocephalus sp والدودة الشريطية للبائن Moniezia expansa وثلاث حزم في الدودة الشريطية للافعى Ophiotaenia europaea وخمس حزم في الدودة الشريطية للطيور Raillietina echinobothrida . | The current research studies cestode tapeworms in four kinds of host vertebrates after dissecting them, including 235 cyprinidae and Barbus Lutus fishes, (15) Hydrophlidia and Microcephalopis gracilis snakes, (25) Loridae and Larus canus seagulls and (95) Ruminant and Ovis ovis .Four system of four kinds of cestde tapeworms were examined using the enzymic method with (AchI) technology. Result show that there is a difference between the central nervous system and the peripheral nervous system. The description for the nervous system for each of these cestode tapworm is : The central nervous system of the Bothriocephalus sp. consist of two pairs of cerebral ganglia; the one large located in the bothria joined other by a cerebral ring, and the other is located in the front part of the scolex at the apical disc, and they are also joined to each other by an apical ring. Both the cerebral ring and the apical ring are linked by a ring nerve commissure. From the large cerebral ganglion and the small ones arise a number of nervous which provide bothria and the front part of scolex.The peripheral nervous system, consist of a main nervous trunk which arise from the big cerebral ganglion, then divides into two pairs of thick lateral trank in each side of the body of the worm, and five lateral nerve cords arise from the apical cerebral ganglion located to the side of the thick nervous trunk, and twenty (20) arises from the big cerebral ganglion. The thick medial nerve cords lateral trunk and the lateral nerve cords with the medial nerve cords are linked by many teraservers commissures. This worm is distinguished by the appearance of a nerve network in every piece of it and expanding toward the exterior edges of the worm body.The central nerve system of the Ophiotaenia europaea consist of a pair of cerebral ganglion located in the area of the suckers which linked with another pairs of apical ganglia smaller than the first pairs located in the apical organ. The apical ganglion and the cerebral ganglia are linked by a ring commissure, and from every ganglia a number of small and thin nerve come out to supply the edge of the scolex, the sucker and the apical organ with nerves.From the cerebral ganglion arise two pairs of the thick lateral nervous suckers, also four pairs of lateral nerve cords raise from the apical ganglion it is noted that there are some nerve commissures between (6 - 8) in every body piece and the existence of thick proglottid from another. This worm is distinguished by the distribution of back lateral nerve ganglion the lateral nerve trunks and on the medial merve cords.The central nervous system of the Raillietina echinobothrida consist of two large cerebral ganglia linked by a circular nerve commissure, and (4 - 6) of small front nerves arise from them to supply the suckers and rostellar muscles. Also from these ganglia two pairs of thick main lateral nerve trunk and four pairs of medial nerve cords arise too. The thick lateral trunk and the medial nerve cords are linked by a number of transvers commissures (6 - 8) ladder nerve commissure, and a thick nerve transverse commissure is noted separating the proglottids from one another, and a nerve degeneration in the area of mature proglottid is also noted.The central nervous system of the Moniezia expansa consist of a pair of cerebral ganglia with many parts called the brain complex linked with each other by a cerebral ring from which a number of thin front of nerves come out to supply the suckers, rostellars and the outer edges of the scolex forming nervous network for the scolex. Also a pair of thick lateral nerve trunks and two pairs of nerve cords, which is characterized decreasing thickness of its lateral trunks.It is noted that the medial nerve cords expand to many proglottids then degenerates when the unripe proglottids starts to conform medial thin nerves and reticulate with the commissures forming a nervous network which ends with ganglion. Interproglottid glands located on the thick transverse commissure which separates the prolottids from one another are also noted, and greatest activity of the acetylcholinestrase enzyme exists in the interproglottid glands.The electronic microscope examination show that the tequment consist of two main layers; external and internal, separated by a layer of muscles, and covered with cytoplasmic projections called microtriches, found to be of three types : filar, conical and figure like, the conical and figure forms appear clearly in (Moneizia expansa), and the basement membrane appear clearly too.The research also include a number of biochemical concentration measuring the total amount of the of the protein, DNA, RNA, carbohydrate, total lipid, cholesterol and enzyme's such as glucose 6 phosphate dehydrogenase activity, Alkaline phosphatase (AIP), Acid phosphatase (AcP), Glutamate pyruvate transaminase (GPT), Glutamate oxaloacetate transaminase (GOT) and Acetylcholinesterase (AchE).The results showed significant differences in the concentration of protein, DNA, RNA, carbohydrate total lipid, cholesterol and enzymes especially in cestode tapeworms that cause bird infection. And also there is a high concentration in total protein that exists in the (Monizia expansa) which causes mammals' infection. It reached to (3.2146  248.667) microgram/cm3. and downed to its lowest levels in birds Raillietina echinobithrida; it reached to (97.000  1.000) microgram to cubicle cm. while the concentration of protein in Ophiotaenia europaea (151.667  2.0817) and (195.000 13.2288) microgram/cm3 in Bothriocephalus sp respectively. Also results shoed the concentration of the DNA was less in Raillietina echinobothrida : (1.000  11.000) microgram/cm3; while it increased to (12.3330.5774) in Ophiotaenia europaea. At the same time the concentration of DNA became in Bothriocephalus sp. and Moniezia expanza (14.3667 0.5508), (24.3332.0817)microgram/cm3. The concentration of the RNA in Moniezia expansa was higher than other types of cestode tapeworms; it reached to (88.6667  3.2146) microgram/cm3. while it reached in Bothriocephalus sp. (67.7133  1.3536), Ophiotaenia europaea (3.055147.333) and Raillietina echinobothrida (23.3332.0817) microgram/cm3.The value of carbohydrate in the cestode tapeworms that cause infection in reptiles was low, while it was higher in those causing sheep infection than other parasites, it reached to (204.5796  2.0801), (186.60009.8989),(291.12671.9688),(329.0008.7178)microgram/cm3 in cestode tapeworms that cause infections in fish, snakes, birds and sheep respectively.The value of total lipids diverged significantly; it was the highest in Moniezia expansa, it reached (229.4270.6839) mg/dl, and (169.05951.2694), (146.20800.7736), and (114.45230.5518) mg/dl in Bothriocephalus sp., Ophiotaenia europea and Raillietina echinobthrida respectively.The study revealed the activity of the enzymes as follows : 1. The Moniezia expansa has a very high activity toward the enzymes : G - 6PHD, AcP, GOT.2. The Bothriocephalus sp. has a higher activity toward the AIP enzymes than other cestode tapeworms, then comes Moniezia expansa in the degree of the activity.3. As for the enzyme GPT the activity was somehow approximate in all the cestode tapeworms.4. The enzyme AchE showed it highest activity in Raillietina echinobothrida then comes Moniezia expansa, while it was low in Ophiotaenia europaea and Bothriocephalus sp in comparison with the two form types.The study of electrophoresis showed different bands in number and thickness; it reached four bands in each type : the one that causes fish infection (Bothriocephalus sp.), and that in mammals (Moniezia expansa) then three bands in Ophiotania europaea, and five bands in Raillietina echinobothrida.

دراسات حول متلازمة المبيض المتعدد الاكياس وتاثير بعض العقاقير المنشطة للخصوبة في مبايض اناث الجرذان == Studies On the Polycystic Ovary Syndrome and Effects Of some Fertility activating Drugs Of Female Rats

Abstract: اجريت الدراسة الحالية التي امتدت من شهر كانون الثاني (2006) لغاية شهر ايار (2008) للتقصي عن انتشار الاصابة بمتلازمة المبيض المتعدد الاكياس لعدد من نساء محافظة نينوى واللاتي بلغ عددهن (92) مصابة، واظهرت الدراسة ان اعلى نسبة للاصابة بحسب الحالات السريرية المرافقة للمتلازمة كانت (51.08 %) في النساء اللاتي يعانين من العقم الاولي وتلتها نسبة الاصابة بالعقم الثانوي (42.39 %) في حين بلغت نسبة الاصابة في النساء الملتحيات (6.52 %). وبينت الدراسة ارتفاع معنوي في تركيز الهورمون المحفز للجريبات ((FSH والهورمون المحفز للجسم الاصفر (LH) مقارنة مع النساء الاصحاء.كما تم تحديد نسبة الاصابة ببعض الامراض التي تعد من المخاطر الصحية في النساء المصابات بالمتلازمة اذ بلغت نسبة الاصابة بارتفاع ضغط الدم (19.56%) في حين كانت نسبة الاصابة بداء السكر(28.26%). اما الاصابة بداء السكر مع ارتفاع ضغط الدم بلغت (5.43%) في النساء المصابات بمتلازمة المبيض المتعدد الاكياس. كما تضمنت الدراسة الحالية متابعة وتشخيص التغيرات النسجية في المبيض المتعدد الاكياس لاناث الجرذان البيضاء البالغة المعرضة للاضاءة المستمرة بشدة (350) لوكس(لمدة 14 اسبوعا) وتمثلت هذه التغيرات بملاحظة مجموعة من التراكيب الكيسية المختلفة الاحجام مع انعدام وجود الجسم الاصفر في المبيض فضلا عن احتقان الاوعية الدموية وازدياد خلوية المبيض. كما شملت الدراسة الحالية متابعة تاثير بعض العقاقير المنشطة للخصوبة المتمثلة بعقاري سترات الكلوميفين بمقدار جرعة (1.6 ) ملغم/ كغم من وزن الجسم لكل دورة شبقية واخرى والميتفورمين بمقدار جرعة (300) ملغم/ كغم من وزن الجسم يوميا كلا على حده وسترات الكلوميفين والميتفورمين معا ولفترات زمنية مختلفة تتراوح بين (4 ، 8 ، 12) اسبوعا من المعاملة بهذه العقاقير ولوحظ ان اعلى زيادة في شدة التغيرات النسجية في مبيض اناث الجرذان عند دمج كل من عقاري سترات الكلوميفين مع الميتفورمين في حين لم تلاحظ تغيرات نسيجية مهمة عند المعاملة بعقار الميتفورمين لوحده ولكن استخدام الكلوميفين لوحده ادى الى حدوث تغيرات نسجية ولكن بشدة متوسطة واقل من دمج العقارين معا وان اهم التغيرات النسجية تمثلت بالتليف وتكيس الجريبات المبيضية مع ازدياد خلوية قشرة ولب المبيض فضلا عن احتقان الاوعية الدموية وزيادة تواجد الاجسام الصفر وهي في مراحل اوبية مختلفة.وشملت الدراسة تقدير تركيز المالوندايالديهايد والكلوتاثايون في نسيج المبيض المتعدد الاكياس لاناث الجرذان البالغة المعرضة للاضاءة المستمرة اذ وجد ان هناك ارتفاعا معنويا في تركيز المالوندايالديهايد مع انخفاض معنوي في تركيز الكلوتاثايون.كما تم دراسة تاثير عقاري سترات الكلوميفين والميتفورمين كلا على حده، وسترات الكلوميفين والميتفورمين معا في تقدير تركيز المالوندايالديهايد والكلوتاثايون في نسيج مبيض اناث الجرذان البالغة ولوحظ ارتفاعا معنويا كبيرا في مستوى تركيز المالوندايالديهايد مع انخفاض معنوي في مستوى تركيز الكلوتاثايون عند دمج العقارين معا. كما لوحظ ارتفاع معنوي ايضا في تركيز المالوندايالديهايد وانخفاض معنوي في تركيز الكلوتاثايون في جميع المعاملات بالعقاقير وازديادها بحسب الفترات الزمنية ما عدا المجاميع المعاملة بعقار الميتفورمين. | The current study, extended from (January 2006 - May 2008) has investigated the infection that spread upon 92 infected women in - Nineveh Province - suffering from polycytic syndrome (PCOS) was conducted. The study showed that the highest rate of infection - according to the associated clinical cases of the syndrome was (51.08%) for women who suffer from primary infertility. Meanwhile, infection percentage for the bearded women was (6.52%). The study also showed a significant increase in the concentrations of Follicle Stimulating Hormone (FSH) and Luteinizing Hormone (LH) compared to those in healthy women. Some health problems were also detected in (PCOS). The hypertension percentage was (19.56%), diabetes (28.26%), while diabetes with hypertension cases were (5.43%) for women who have (PCOS). The present study also involved monitoring and diagnosis of tissue changes in polycytic ovary among white adult female rats, which were exposed to continuous light with an intensity of 350 lux (for 14 weeks). Those changes were represented by identifying a number of cystic structures of various sizes, the absence of corpus luteum, congestion of blood vessels and ovarian hyperplasia. The study also investigated the influence of fertility - activating drugs, like clomiphene citrate (each dose is 1.6 mg/ kg of body weight for one dose estrus stage), metformin (each dose is 300 mg/ kg of body weight daily). Each of the drugs was used alone and clomiphene citrate was associated with metformin were given for different periods (4, 8, and 12) weeks. The highest change in the ovary tissues of female rats was found when using clomiphene citrate with metformin drugs. While no significant tissue changes were observed when using metformine alone. But the treatment by using clomiphene citrate alone resulted in medium tissue changes, and were less when combining the two drugs together. The most important changes are fibriosis ovarian follicle cysts, hyperplasia in the cortex and the ovary mudella as well as the congestion of blood vessels in addition to an increase in the number of corpus luteum when they are in different aterisa stages. The study also included the estimation of the concentration of malondialdehyde and glutathion in the polycystic ovary tissue of adult female rats exposed to continuous light. Its was found that there was a significant increase in the concentration of malondialdehyde and a significant decrease in the concentration of glutathion. In addition to that, the influence of clomiphene citrate of metformin drugs separately, or together, to estimate the concentrations of malondialdehyde and glutathion in adult female ovary tissues was also investigated. Malondialdehyde concentration was high, while glutathion concentration was low when combining the two drugs. Also, a higher significant increase was found in the malonaldehyde concentration and a significant decrease in glutathaion concentration in all drug treatments and their increase over different periods of time, except for the groups, which were treated with metformin.

Abstract: اجريت دراسات حياتية وبيئية لحشرة من القطن Aphis gossypii Glover ( Homoptera : Aphididae ) التي تعد من الافات الاقتصادية عالمية الانتشار، والتي تصيب العديد من النباتات منها اشجارو محاصيل الخضر بانواعها فضلا عن نباتات الزينة والنباتات البرية . شملت الدراسة ما يا تي : اولا : الدراسات المختبرية ا - تناسبت مدة الاطوار الحورية عكسيا مع درجات الحرارة اذ كانت اقصر مدة للطور الحوري الاول ( 1.25 ) يوم في حرارة ( 30 ) م° ، واطول مدة لهذا الطور كانت في حرارة ( 15 ) م° ( 4.4 ) يوم . تراوحت مدة الطور الحوري الثاني بين ( 4.1 - 1.2 ) يوم في درجتي الحرارة ( 15 , 30 ) م° . اما مدة الطور الحوري الثالث فكانت متدرجة بين ( 3.5 - 1.1 ) يوما في الحرارة ( 15 , 30 ) م° ، اما مدة الطور الحوري الرابع فكانت اقل مدة في حرارة ( 30 ) م° اذ بلغت ( 1.05 ) يوم واطول مدة كانت ( 3.4 ) يوم في حرارة ( 15 ) م° . كانت اطول مدة للدور الحوري ( 15.4 ) يوم في حرارة ( 15 ) م° واقصرها ( 4.6 ) يوم في حرارة ( 30 ) م° . بلغ اعلى انتاجية للانثى العذرية ( 54.1 ) حورية في حرارة ( 25 ) م° ، واقل انتاجية كان في ( 15 م°) . كما وجد ان طول مدة قبل التكاثر تتاثر ايضا بدرجات الحرارة اذ اطول مدة كانت ( 3.2 ) يوم في حرارة ( 15 ) م° واقصر مدة كانت ( 0.9 ) يوم في حرارة ( 30 ) م° . كما ان مدة التكاثر تتاثر ايضا بدرجات الحرارة اذ وجد ان اطول مدة كانت ( 11.2 ) يوم في حرارة ( 15 ) م° واقل مدة ( 6.2 ) يوما في حرارة ( 30 ) م° . بلغ معدل انتاج الانثى حورية الواحدة في اليوم الواحد اعلاه ( 6.88 ) حورية / انثى / يوم في حرارة ( 25 ) مº ، واقلها ( 1.61 )حورية / انثى / يوم في ( 15 ) مº . ان طول عمر الانثى يتناقص بارتفاع درجة الحرارة اذ كان اطول عمرا ( 41.3 ) يوم في حرارة ( 15 ) مº ، واقل عمرا ( 19.9 ) يوم في حرارة ( 30 ) مº . وجد ان مدة الجيل كانت تتناسب عكسيا مع درجات الحرارة ، وكانت اطول مدة ( 18.6) يوم في حرارة ( 15 ) م° واقصر مدة كانت ( 5.5 ) يوم في حرارة ( 30 ) م° .ازدادت النسبة المئوية للهلاك براتفاع الحرارة من 20م° الى ان وصلت اقصاها 100 % في حرارة 35م°. ب - ان لدرجة الحرارة ونوع المضيف النباتي تاثيرا واضحا في مختلف الجوانب الحياتية للحشرة فلوحظ التاثير كان كبيرا في مدة الدور الحوري والانتاجية وطول العمر ومدة الجيل والنسبة المئوية للهلاكات على المضائف النباتية المختلفة . ثانيا : الدراسات الميدانيةا ــ المضائف النباتية : وجد من المسح الميداني للمضائف النباتية التي تصيبها حشرة من القطن في حقول الجادرية انها تصيب اكثر من 40 نوعا من 11 عائلة نباتية اغلبها من العائلات : القثائية ، والمركبة ، والباذنجانية ، والخبازية . ب ــ الكثافة العددية : تختلف باختلاف المضائف النباتية اذ وجد ان اكثر النباتات اصابة هي البطيخ ،و خيارماء ثم الباميا في خريف 2005 . اما في سنة 2006 وجد ايضا ان الكثافة مختلفة باختلاف المضائف ، وان اعلى كثافة على البطيخ والخيارو القطن ثم الباميا ، كما انها تختلف من اسبوع لاخر باختلاف درجات الحرارة والرطوبة النسبية ج ــ تفضيل الحشرة لسطحي الورقة : ان اعلى كثافة لحوريات من القطن كانت على السطح السفلي ( 370.4 ، 195.2 ، 464.5 ) لورقة نبات خيار الماء ، والباذنجان ، والباميا على التوالي واقلها على السطح العلوي ( 132.7 ، 54.0 ، 60.1 ) على التوالي وان اعلى نسبة مجنحات على السطح السفلي على النباتات الثلاث . د ـ توزيع الحشرة على مستويات النبات الثلاث : وجد ان اعلى نسبة للافراد غير المجنحة على المستوى الوسطي ثم القاعدي ثم القمي على نباتات خيار ماء ، وباذنجان ، وباميا على التوالي . اما بالنسبة للمجنحات فكانت مختلفة باختلاف النباتات على السطح العلوي للورقة . اما على السطح السفلي فوجد ان اعلى معدل للافرا د غير المجنحة كان على المستوى الوسطي ثم القاعدي ثم القمي للنباتات الثلاث ، على التوالي في حين انتشار الافراد المجنحة يختلف باختلاف النباتات . هـ - ان توزيع الحوريات غير المجنحة على نصل الورقة كان اعلاه على المساحات الخضر مابين العروق ، واقلها على العرق الوسطي للسطح السفلي لورقة المضيف النباتي . | The cotton aphid , Aphis gossypii Glover ( Homoptera : Aphididae ) is an important economic cosmopolitan polyphagous pests Which attacks numerous hosts such as trees , field crops , ornamental and wild plants . The biology of this insect has not been adequately studied in Iraq .Hence ,this investigation was designed to study the local strain of the pest under controlled and field condition .These investigation covers the following aspects : First : Laboratory studies : 1 - The nymphal stadia decrease with increasing temperatures . The first stadium reneged from 1.25 days at 30 C° to 4.4 days at 15 C° . Second stadium ranged between 4.1 - 1.2 days at 15 - 30 C°, The third stadium was from 3.5 to 1.1 days at 15 - 30 C°, whereas the fourth stadium was 1.05 - 3.4 days at 30 C°, 15C°. Temperature 35 C° is considered a lethal degree of most nymphal instars .The total period of nymphal stage was 15.4 days at 15 C° and 4.6 days at 30 C°.fecundity value The highest of apterae was 54.1 nymphs at 25 C° and the lowest at 15 C° .The maximum daily reproductive rate of a viviparous female was 6.88 nymphs / female /day at 25 C° and the minimum was 1.61 nymphs / female /day at 15C° . Prereproductive and , reproductive period was also affected with temperature .Generation period from birth to birth ranged between 20.9 days at 15 C° to 7.3 days at 30 C°. The maximum longevity was 41.3 days at 15C° and the minimum was 19.9 days at 30 C° . 2 - The laboratory studies have generally revealed that the degree of temperature and host plants have significant effects on the bionomics of cotton aphid . Second : Field studies Cotton aphid has a wide range of host plants . The field survey shows that more than 40 species of host plants belong to the families : Compositae , Malvaceae , Cucurbitaceae and Solanaceae in Jadriyah fields Population density varies on different host plants . The aphid prefers melon , cucumber , okra and cotton and its population varies depending on temperature , relative humidity and natural enemies . The field studies have revealed that both alatae and apterae of cotton aphid prefer the lower surface of different host plant leaves . The highest distribution of them was on the middle leaves of plant and high percentage ratio of apterous on the green area while the lesser one was on the midrib of leaves

Abstract: عني البحث باستخدام خمسة جالكونات مصنعة ( 1،3 - ثنائي الاريل - 2 - بروبين - 1 - ون ) كمواد بادئة لتصنيع مختلف المركبات الحلقية غير المتجانسة. اختبرت بعدها الفعالية الحيوية للمركبات المصنعةالمذكورة ضد امامية السوط promastigotes لطفيلي اللشمانيا الاستوائية Leishmania tropica. في الزجاج .in vitroكثفت الجالكونات الخمسة المصنعة مع مركبات ذات هيدروجين حامضية Acidic Hydrogen compounds مثل الهايدرازينات Hydrazines واليوريا Ureas ، والثايوريا Thioureas لانتاج خمسة من المركبات النهائية. اثنين من تلك المركبات احتوت حلقة بايرازولين وهي : 5,1 - ثنائي فنيل - 3 - (اورثوكلوروروفينيل) - 2Δ - بايرازولين (P1) و3 - (ميتانيتروفنيل) - 5,1 - (باراكلوروفنيل) - 2Δ - بايرازولين (P5), المركبات الثلاثة الاخرى احتوت حلقة بيرميدينون وهي : 4,3 - ثنائي الهيدروجين - 4 - (بارانايتروفينيل) - 6 - فينيل بيرميدين - 2(H1) - ون(P2) و4,1 - ثنائي الهيدروجين - 3 - اثيل - 4 - (2 - مستيلينيل) - 6 - (اورثوكلوروفينيل) بيرميدين - 2(H1) - ون (P4) و2،4 - ثنائي الهيدروجين - 4 - (باراميثوكسي فينيل) - 6 - (اورثوكلوروفينيل) بيرميدين - 2 - (H1) - ثايون (P3) ، والحاوي على حلقة بيرميدين ثايون . وشخصت المركبات المحضرة باستخدام الطرق الطيفية . اختبرت فعالية الحلقات الخمسة متباينة ضد امامية السوط لطفيليات اللشمانيا الاستوائية في الزجاج In vitro. تبين ان المواد الخمسة المختبرة كان لها تاثير تثبيطي على نمو امامية السوط، وازداد هذا التاثير مع زيادة تركيز المادة قيد الاختبار.عندما قورن تركيز عقار البنتوستام Pentostam المثبط لخمسين بالمئة (IC50) من امامية السوط (20مايكروغرام/ملليلتر ) مع IC50 للمواد قيد الدراسة، وجد ان كل المواد المجربة سببت تثبيطا جيدا(نسبيا) في نمو الطفيلي؛ وان اكثر المواد سمية" كان المركب P4 حيث ان IC50 له كانت (0.6 مايكروغرام/ملليتر) ,بينما امتلك المركب P3 IC50 مماثل لعقار البنتوستام (20 مايكروغرام/ملليلتر) اما المركب P5 فقد امتلك IC50 اعلى بقليل من البنتوستام ( 25مايكرو غرام/ملليلتر) . وكانت IC50 للمركب P1 (30مايكروغرام/ملليلتر). بينما كانت IC50 للمركب P2 (60مايكروغرام/ملليلتر) وهو الاقل سمية" على امامية السوط من بين المركبات الخمسة.درس تاثير المركبات الخمسة على ايض الطاقة في امامي السوط للشمانيا الاستوائية من حيث التاثير على محتوى الاحماض الدهنية وعلى بعض انزيمات السلسلة التنفسية المعلوم تواجدها في امامي السوط.حللت الاحماض الدهنية في امامي السوط للشمانيا الاستوائية بالاعتماد على تقنية كروماتوغرافيا الغاز السائل (Gas liquid chromatography) وتبين وجود ستة انواع من الاحماض الدهنية في امامي السوط وهي على التوالي : Caproic acidو Myristic acidو Palmatic acidو Stearic acidوOleic acid وLinoleic acid.اعطى IC50 للمواد الخمسة (P5 - - - P1) تاثيرات كمية ونوعية متباينة على محتوى امامية السوط من الاحماض الدهنية، فقد اعطى IC50 للمركب P3 اقوى تاثير على محتوى الاحماض الدهنية حيث سبب اختفاء اغلب انواع الاحماض الدهنية ،مع زيادة ملحوظة في نسبة تركيز Caproic acid (35%) ، كما تسبب IC50 للمركب P4 في اختفاء Oleic acid لامامية السوط المعاملة، مع نقص ملحوظ في نسبة تركيز Linoleic acid. وعند معاملة امامية السوط ب IC50 للمركب P5 حصل نقص كبير في نسبة تركيز oleic acid وLinoleic acid. على ما يبدو، فان Linoleic acid وOleic acid هما اكثر الاحماض الدهنية تاثرا، حيث انهما فقدا كليا عند معاملة امامي السوط بـ IC50 للمركبات P3و P4وP5.اختبر تاثير IC50 للمركبات (P5 - - - P1) على فعالية انزيمي فيوماريت ردكتنيز ((FRD وسكسينت ديهيدروجينيز ((SDH. وتبين ان اربعة من المركبات المصنعة وهي P2 - - - P5 امتلكت تاثيرا انتقائيا على السلسلة التنفسية لامامية السوط، حيث اثرت المواد انفة الذكر على فعالية FRD اكثر من تاثيرها على فعالية SDH. وكان تاثير المركب P2 على فعالية FRD اكثر بنسبة 34% من تاثيره على فعالية SDH، في حين كان تاثير المركب P3 على فعالية FRD اكثر ب54% من تاثيره على فعالية SDH. وكذلك الحال بالنسبة للمركب P5 الذي ثبط فعالية FRD بنسبة 43% اكثر من تثبيطه لفعالية انزيم SDH. تشير هذه البيانات الى ان انزيم FRD للطفيلي يمكن ان يكون الموقع المستهدف (الهدف الخاص) الذي يعكس فعالية المركبات الثلاثة P2و P3 وP5 المضادة لامامية السوط للشمانيا الاستوائية. بينت الدراسة الحالية ان المركبات P5 - - - P1 تسببت باختزال المحتوى الكلي للبروتينات والاحماض النووية وكذلك اثرت على فعالية بعض الانزيمات المتعلقة بايض هذه الجزيئات وهما انزيمي دايهايدروفوليت ردكتيز (DHFR) وثايميدين فوسفوريليز (TP). اظهر IC50 للمركب P5 اقصى اختزال في المحتوى البروتيني لامامية السوط (35%) وتسبب كل من IC50 للمركبات P1و P3وP4 باختزال في المحتوى البروتيني لامامية السوط بدرجات اقل : (27%، 24%، 19%على التوالي)، اما IC50 للمركب P2 فقد كان الاقل تاثيرا على محتوى البروتين حيث ادى الى اختزال 9% فقط من محتوى البروتين الكلي. اما من حيث التاثير على محتوى الاحماض النووية لامامية السوط المعاملة؛ فقد سبب P5 اقصى اختزال في كمية الاحماض النووية الكلية (42%). كما تسبب P2و P3و P4 باختزال محتوى الاحماض النووية لامامية السوط بنسب متقاربة (35%،36%،37%على التوالي )، في حين سبب P1 اقل اختزال بمحتوى الاحماض النووية(16%). بينما اعطى P5 اعلى اختزال في كمية الحامض النووي الرايبوزي منقوص الاوكسجين ( DNA )(50%) في حين ان P1 سبب اقل اختزال في كميةDNA (12%) . واما ما يخص محتوى امامية السوط من الحامض النووي الرايبوزي (RNA )، فقد اختزل المركب P4 41% من RNA الكلية لامامية السوط المعاملة واختزل IC50 للمركب P5 39% من RNA لامامية السوط المعاملة. وسجل اقل اختزال في محتوىRNA عند معاملة امامية السوط ب IC50 للمركب P1 (17%). تبين ان المركبات قيد الدراسة اثرت على كل من مساري البناء الحيوي de novo والانقاذ salvage لبناء dTMP. حيث ان هذه المواد (P5 - - - P1 - ) اثرت معنويا في فعالية انزيم TP . فالمركبات P2و P3و P4و P5 اختزلت الفعالية النوعية لانزيم TP في امامية السوط، من ناحية اخرى فان جميع المركبات قيد الدراسة تسببت في اختزال الفعالية النوعية لانزيم DHFR باستثناء المركب P1 الذي تسبب في زيادة الفعالية النوعية لانزيم DHFR الى 106%. وان اقصى اختزال في فعالية DHFR سببه المركب P3 (34%).يجدر بالذكر ان IC50 للمواد قيد الدراسة كان لها تاثيرا ملحوظا في شكل وابعاد الطفيلي. حيث تسبب IC50 للمركب P5 بظهور فجوات كبيرة في السايتوبلازم، اما المركب P2 فقد تسبب في ظهور تحبب في السايتوبلازم مع نواة غير منتظمة الشكل. سببت معاملة امامي السوط ب IC50 للمركب P3 ظهور تجمع من الفجوات السايتوبلازمية الصغيرة، من ناحية اخرى ظهرت فجوة منفردة كبيرة في سايتوبلازم الطفيليات المعاملة بالمركب P4، مع نواة غير منتظمة الشكل. لوحظ اقصى انتفاخ في امامية السوط المعاملة بـ IC50 للمركب P4 وبدرجات اقل لتلك المعاملة بـ اولية IC50 للمركبات P1و P2 وP5 على التوالي.بينت نتائج البحث الحالي ان موقع ونوع المجاميع التعويضية على الحلقات المصنعة في المركبات قيد الدراسة، كانت ذا تاثير حرج في الفعالية المضادة للشمانيا لهذه المركبات. | The present investigation is concerned with the use of five synthesized chalcones (1,3 - diaryl - 2 - propen - 1 - one) as a chief starting materials to synthesize different heterocyclic compounds. The produced heterocycles tested then for their biological activity against Leishmania tropica promastigotes in vitro.The five synthesized chalcones were condensed with different acidic hydrogen compounds such as hydrazines, ureas and thioureas to produce five heterocyclic compounds. Two of them contain pyrazoline ring : 1,5 - Diphenyl - 3 - (o - chlorophenyl) - ∆2 - pyrazoline (P1) and 3 - (m - Nitropheny - 5 - (p - chlorophenyl) - ∆2 - pyrazoline (P5), the other three compounds contain pyrimidinone ring : 3,4 - Dihydro - 4 - (p - nitro phenyl) - 6 - phenyl pyrimidine - 2(1H) - one (P2), 1,4 - Dihydro - 3 - ethyl - 4 - (2 - mesitylenyl) - 6 - (o - chlorophenyl) pyrimidine - 2(1H) - one(P4),and,4 - Dihydro - 4 - (p - methoxyphenyl) - 6 - (o - chlorophenyl) pyrimidine - 2 - (1H) - thione (P3), which contain pyrimidine thione ring. The resultant products were identified in the light of spectroscopic methods, as well as the melting points. The five heterocyclic compounds were tested for their antileishmanial activity against Leishmania tropica promastigotes in vitro. All tested compounds caused concentration - dependent inhibition of the in vitro growth of Leishmania promastigotes. When compared with inhibitory concentration 50 ( IC50 ) of pentostam (20μg/ml), all the tested compounds appeared to have relatively good inhibitory effects. The IC50 values of the tested compounds P1,P2,P3,P4 and P5 were 30 μg/ml, 60 μg/ml, 20 μg/ml, 0.6 μg/ml and 25 μg/ml respectively.Effects of the five synthesized compound were also tested on energy metabolism (on fatty acids content and some selective enzymes in respiratory chain) of Leishmania promastigotes. The gas liquid chromatographic analysis of untreated Leishmania tropica promastigotes revealed the presence of six types of fatty acids : caproic, myristic, palmatic, stearic, oleic, and linoleic acids respectively. The IC50 of the tested products (P1 - - - P5) showed different effects, qualitative and quantitative on the fatty acids content of promastigotes. IC50 of P3 displayed the maximum effect on promastigotes fatty acids content, in which most types of fatty acids disappeared, with considerable increase in concentration percentage of caproic acid (35%). IC50 of P4 was caused oleic acid of the treated promastigotes to disappear, with remarkable decrease in percentage concentration of linoleic acid. When promastigotes treated with IC50 of P5, there was a high decrease in percentage concentration of oleic acid and linoleic acid respectively. Oleic acid and linoleic acid seemed to be the most affected fatty acids, since they were missed in the promastigotes when treated with P3, P4. and P5. Fumarate reductase (FRD) and succinate dehydrogenase (SDH) were investigated for its sensitivity toward the five compounds under study (P1 - - - P5). IC50S of four experimented compounds (P2 - - - P5) was observed to have selective inhibitory effects on the parasite respiratory chain, since these compounds proved to have higher inhibitory effects against FRD activity than that against SDH activity. However, the inhibitory effect of P2 on the activity of FRD was approximately 34% higher than that on SDH, while the inhibitory effect of P3 on the activity of FRD was 54% higher than that on SDH, and the inhibitory effect of P5 on the activity of FRD was approximately 43% higher than that on SDH i.e. FRD is more sensitive to P2, P3 and P5 than SDH. The data indicate that the parasite FRD might be the specific target for the action of P2, P3 and P5.IC50 of P1 - - - P5 appeared to have variant inhibitory effects on the total protein, total nucleic acids content and some related enzymes (dihydrofolate reductase (DHFR) and thymidine phosphorelase (TP)). P5 caused the maximum decrease in protein content (35%); P3, P4 and P2 reduced protein total content of promastigotes for less extent (27%, 24%, and 19% respectively); P2 resulted in the less reduction in total protein content (9%). As for nucleic acids content of treated Leishmania promastigotes, the maximum reduction of nucleic acids content caused by P5 (42%), P2, P3, P4 caused approximate reduction percentage in total nucleic acids (35%, 36%, 37% respectively), while P1 resulted in the less inhibition of nucleic acids content (16%). The maximal inhibition in Deoxyribonucleic acid (DNA) content caused by P5 (50%), whereas P1 caused the minimal inhibition (12%). However, Ribonucleic acid (RNA) content was reduced 41% when Leishmania promastigotes treated with P4, and 39% when treated with P5. The less reduction in RNA content was caused by P1 (17%). The experimented compounds appeared to affect both salvage and de novo pathways of Deoxy Thymidinemonophosphate (dTMP) synthesis. However all these compounds (P1 - - - P5) resulted in significant changes in the activity of promastigotes TP. P2, P3, P4 and P5 reduced the specific activity of promastigotes Thymidine phosphorelase (TP). On the other hand, all tested products had inhibitory effects against DHFR activity, except P1, which increases the activity of promastigotes Dihydrofolate reductase (DHFR) to 106%. P3 has the highest inhibitory effect on promastigotes DHFR - activity, since it reduced DHFR activity to 34%. Inhibitory concentration 50 ( IC50 ) of the experimented compounds made remarkable changes in the morphology and dimensions of the parasite. Large vacuoles in the cytoplasm were observed in the parasites, which were treated with IC50 of P1. Cytoplasmic granulation and amorphous nuclei were observed in the parasites, which were treated with IC50 of P2. Assemblage of small vacuoles was observed in the parasites that were treated with IC50 of P3. On the other hand, parasites that were treated with IC50 of P4 appeared to contain single large vacuole that occupies large space of the cytoplasm, with amorphous nuclei. Maximum swell was observed in the promastigotes that were treated with IC50 of P4 and at lower extent with those treated with IC50 of P1, P5, and P2 respectively. The results of the present research showed that the position and type of the substituent on the synthesized rings of the tested products seemed to be critical for their antileishmanial activity.

Abstract: يمتاز مرض قصور الدرقية بانخفاض مستوى هرمونات T3 وT4 الضرورية لعمليات انتاج الطاقة وحيوية الجسم ؛ لذلك ترافق هذه الحالة اضطرابات في عمليات الايض المختلفة في الجسم . اجريت الدراسة في المركز التخصصي للغدد الصم والسكري في بغداد ، وشملت 70 مريضا من مرضى قصور الدرقية ، تم تشخيص الحالة لديهم عن طريق قياس مستويات الهرمونات (TSH,T4,T3) . كما وشملت 60 شخصا اختيروا بشكل عشوائي من غير مرضى الغدة ، وقد تم التاكد من سلامة الغدة وعدم وجود قصور في وظيفتها لديهم. تضمنت الدراسة الكشف عن نسبة حدوث مقاومة الانسولين لدى مرضى الغدة ومقارنتها مع نسبة حدوثها لدى الافراد الاصحاء الغدة. وقد تم تشخيص مقاومة الانسولين باستخدام اختبارات (الببتيد الرابط C - peptide واختبار تحمل الكلوكوز الفموي Oral glucose tolerance test (OGTT) ، اذ ان ارتفاع مستوىCP والتحمل الضعيف للكلوكوز Impaired glucose tolerance (IGT) يشيران الى مقاومة مؤكدة للانسولين Definite insulin resistance (DIR) ، في حين وجود اضطراب في نتيجة احدهما يشير الى مقاومة ممكنة للانسولين Possible insulin resistance (PIR) . تشير نتائج الدراسة الحالية الى ان كلتا الحالتين المرضيتين (القصور الدرقي ومقاومة الانسولين) مترافقتان وشائعتان لدى النساء وكبار السن وكذلك لدى الافراد من فئة المفرطي الوزن (BMI=25 - 29.9 kg/m2) والبدناء(BMI>30 kg/m2) ، فقد وجد ان اغلب مرضى قصور الدرقية المؤكدي المقاومة للانسولين هم من الفئات العمرية 40 - 21 سنة و60 - 41 سنة وقد شكلوا نسبة %38.5 لكل فئة وكان اغلبهم من النساء ، وشكلوا نسبة %84.6 بالمقارنة مع نسبة %15.4 للذكور . اما المرضى الممكني المقاومة للانسولين فكان اغلبهم من الفئة العمرية 60 - 41 سنة وشكلوا نسبة %66.7 واغلبهم من النساء ايضا وشكلوا نسبة %77.8 بالمقارنة مع نسبة %22.2 للذكور . كذلك يلاحظ ان اغلب المرضى المؤكدي المقاومة للانسولين هم من فئة البدناء وشكلوا نسبة %53.8 واغلب المرضى الممكني المقاومة للانسولين هم من فئة المفرطي الوزن وشكلوا نسبة %51.9 . وجد من نتائج البحث ارتفاع نسبة حدوث مقاومة الانسولين لدى مرضى قصور الدرقية ، فقد وجد ان %18.6 منهم لديه مقاومة مؤكدة للانسولين ، في حين كانت النسبة اقل بلغت %3.3 لدى الافراد الاصحاء الغدة . وبلغت نسبة المرضى الذين لديهم مقاومة ممكنة للانسولين %38.6 بالمقارنة مع نسبة %25 للافراد الاصحاء . وهي نسبة مرتفعة تؤكد دور قصور وظيفة الغدة الدرقية في نشوء مقاومة الانسولين. تضمنت الدراسة كذلك قياس مستويات بعض المؤشرات البايوكيميائية والمناعية لدى مرضى قصور الدرقية المؤكدي او الممكني المقاومة للانسولين ومقارنتها مع مستوياتها لدى افراد السيطرة الاصحاء. وشملت هذه المؤشرات قياس مستوى الكلوكوز في حالة الصيام ، وCP ، وانواع الدهون ، واختبار OGTT ، وHbA1c ، وTNF - α ، وIL - 6 ، وCRP ، والنسبة المئوية لذوي الخلايا اللمفية في الدم المحيطي . اظهر مرضى قصور الدرقية الذين لديهم مقاومة مؤكدة للانسولين تحملا ضعيفا للكلوكوز Impaired glucose tolerance (IGT) وذلك من نتائج اختبار OGTT ، وكذلك ارتفاعا في مستوى الكلوكوز في حالة الصيام بلغ 1.24±5.77 مليمول/ لتر ، وارتفاع مستوى CP بلغ 1.71 ±5.39 نانوغرام/ مليلتر ، وارتفاع معنوي في مستوى HbA1c والبالغ 0.84± %5.86 ، وقد كان مستوى هذه المؤشرات لدى هذه المجموعة اعلى من مستوياتها لدى المجموعتين الاخريين (NIR,PIR) وبالمقارنة مع السيطرة . شملت مجموعة مرضى قصور الدرقية الممكنة المقاومة للانسولين %10 لديهم مستوى مرتفع لـ CP و%28.6 لديهم تحمل للكلوكوز ضعيف (IGT) ، وبشكل عام اظهرت هذه المجموعة ارتفاعا معنويا عاليا في مستوى الكلوكوز في حالة الصيام بلغ 1.97±5.63 مليمول/ لتر وارتفاعا معنويا عاليا (P<0.01) في مستوى CP ، اذ بلغ 2.82±3.83 نانوغرام/ مليلتر ، وارتفاعا معنويا عاليا (P<0.01) في مستوى HbA1c والبالغ 1.62±%6.21 . اما بالنسبة لاختبار ذوي الخلايا اللمفية في الدم المحيطي ، فقد لوحظ ارتفاع معنوي عال في نسبتها لدى مجاميع المرضى المقاومة للانسولين مؤكدة او ممكنة بالمقارنة مع مجموعة السيطرة. اوضحت دراسة المؤشرات المناعية وجود نمط التهابي تحت سريري لدى مرضى قصور الدرقية المؤكدي المقاومة للانسولين ، وذلك من الارتفاع المعنوي العالي(P<0.01) لمستويات كل من TNF - α وIL - 6 التي بلغت 1.02± 6.46 بيكوغرام / مليلتر و2.14 ±50.6 بيكوغرام / مليلتر على التوالي بالمقارنة مع مجموعة السيطرة. اما مستوى البروتين الفعال سي C - reactive protein (CRP) فقد لوحظ ارتفاع مستوياته لدى مرضى قصور الدرقية جميعا سواء كانوا من المقاومين ام غير المقاومين للانسولين ، مما يشير الى كونه عاملا التهابيا غير نوعي له ارتباط بمرض قصور الدرقية ولكن ليس له دور في التفريق بين الحالات المقاومة وغير المقاومة للانسولين . اظهرت دراسة العلاقات الترابطية بين المؤشرات البايوكيميائية والمناعية التي تم قياسها لدى مجاميع الدراسة وجود علاقة ترابطية سالبة معنوية (P<0.05) بين CP وكل من HbA1c وT.Ch وHDL - C وF.G (P<0.01) . كذلك وجدت علاقة ارتباطية ايجابية معنوية عالية (P<0.01) بين TNF - α وكل من T.Ch وLDL - C ، وعلاقة ارتباط سالبة معنوية عالية (P<0.01) بين TNF - αو TSH لدى مجموعة المرضى المؤكدي المقاومة للانسولين . وفي مجموعة المرضى الممكني المقاومة للانسولين لوحظ وجود علاقة ارتباطية ايجابية معنوية (P<0.05) بين مستوى IL - 6 وBMI وF.G و(P<0.01) HbA1c . اما بالنسبة للبروتين الفعال سي فقد لوحظ وجود علاقة ارتباطية سالبة معنوية (P<0.05) بين مستواه ومستوى T.Ch وعلاقة ارتباطية ايجابية معنوية (P<0.05) بين CRP وكل من F.G وCP لدى مجموعة المرضى المؤكدي المقاومة للانسولين و(P<0.01) TNF - α لدى مجموعة المرضى الممكني المقاومة للانسولين. اظهرت الدراسة ارتفاع العامل الالتهابي IL - 6 لدى المرضى المؤكدي المقاومة للانسولين ونظرا لدوره المهم في العمليات الايضية المختلفة في الجسم ؛ لذلك وجد انه من الضروري دراسة تاثيره في وظيفة الغدة الدرقية وفعاليات اخرى ومقارنته مع تاثير عقار الكاربيمازول . وقد تم ذلك عن طريق حقن ثلاث مجاميع من الجرذان ، الاولى بالانترلوكين - 6 ، والثانية بعقار الكاربيمازول ، والثالثة بالمحلول الملحي الفسيولوجي بوصفها سيطرة. بينت هذه التجربة ان كل من الانترلوكين - 6 والكاربيمازول يسببان انخفاض مستويات هرمونات الدرقية (T4,T3) عند حقنها في الحيوانات ، كما ويسببان انخفاضا معنويا في وزن الجسم في حين لم يكن لهما تاثير في وزن الكبد. لوحظ انخفاض مستوى الكلوكوز في الدم لدى الحيوانات المحقونة بعقار الكاربيمازول ، بينما لم يحدث تغير في مستواه بتاثير الانترلوكين - 6 . اما بالنسبة لانواع الدهون فقد لوحظ ارتفاع معنوي لمستوى كل من T.G وVLDL - C لدى الحيوانات المعاملة بالانترلوكين - 6 ، في حين لم تتاثر مستويات الدهون في الحيوانات المعاملة بالكاربيمازول. واخيرا ادى حقن الحيوانات بالانترلوكين - 6 والكاربيمازول الى زيادة ذوي الخلايا اللمفية في الدم المحيطي . | Hypothyroidism is a condition in which thyroid hormones levels decreased in the blood. These hormones are necessary for energy production and body viability. In many occasions this condition is accompanied or followed by different metabolic disorders. The current study is conducted in the "Specialized center for endocrinology and diabetes" and carried on 70 hypothyroid patients and 60 randomly chosen individuals with normal thyroid function .Both groups were submitted to laboratory tests to evaluate thyroid function (T3,T4.TSH). The study involved evaluation of the relationship between hypothyroidism and insulin resistance (IR) . Health problem related to many diseases , became common lately. Insulin resistance diagnosed through two important tests : C - peptide (CP) and oral glucose tolerance test (OGTT). Patients are considered to have definite insulin resistance (DIR), if they have impaired glucose tolerance (IGT) and high level of CP. If one of the tests is abnormal, the patients have possible insulin resistance (PIR). The current study show that both conditions (hypothyroidism and insulin resistance) are associated and common in women, old and overweight individuals. The results show that most hypothyroid patients with DIR are at age between 21 - 40 and 41 - 60 year, which constitute about 38.5% for each. Most of them are women constituting 84.6% compared with 15.4% for men. Most hypothyroid patients with PIR are in the ages between 41 - 60 year, constituting 66.7%. The women constitute about 77.8% compared with less percentage for men 22.2%. Also we should notice that most definite insulin resistant hypothyroid patients are obese , and constitute about 53.8% , and most possible insulin resistant hypothyroid patients are overweight, and constitute about 51.9% Insulin resistance incidence is increased in hypothyroid patients, the results show that 18.6% of hypothyroid patients have DIR compared with less percentage 3.3% in healthy individuals , and about 38.6% hypothyroid patients have PIR compared with 2.5% in healthy individuals This study also considered some biochemical and immunological parameters which include : Fasting blood glucose, (FBG), CP, Lipid profile, OGTT, HbA1c, TNF - α, IL - 6, CRP and percentage of lymphocytes apoptosis in peripheral blood. The levels of these parameters are compared between the study groups. Hypothyroid patients with DIR show impaired glucose tolerance (IGT) and high level of fasting glucose reaching 5.77±1.24 mM/l , high level of CP reaching 5.39±1.71 ng/ml and high level of HbA1c reaching 5.86% ±0.84 .The values of these parameters in this group are higher when compared with other groups and control. About 10% of hypothyroid patients with PIR have high level of CP and 28.6% have IGT. In general, PIR hypothyroid patients show a significant (P<0.01) increase in F.G reaching 5.63±1.97 mM/l , significant (P<0.01) increase in CP level reaching 3.83 ±2.82 ng/ml , and significant increase in HbA1c reaching 6.21% ±1.62. The results of peripheral lymphocytes apoptosis test show significant (P<0.01) increase in hypothyroid patients (DIR or PIR) compared to control group. The results show a significant (P<0.01) increase in levels of TNF - α reaching 6.46±1.02 pg/ml and 50.6±2.14 pg/ml respectively , in hypothyroid patients with DIR compared to control and the high levels of these parameters refer to subclinical inflammation . The non - specific inflammatory marker CRP , which refers to systemic inflammation , also increases in all hypothyroid patients (Insulin resistant or none) The current study also includes the study of correlations between. biochemical and immunological parameters in the study groups. The results show that there was a negative significant correlation between CP and each of : HbA1c, T.G, HDL - C (P<0.05) and F.G (P<0.01). There was a positive significant correlation between TNF - α and each of T.Ch, LDL - C (P<0.01) and negative significant correlation between TNF - α and TSH (P<0.01) in DIR hypothyroid patients. Patients with PIR show positive significant correlation between levels of IL - 6 and each of : BMI, F.G (P<0.05) and HbA1c (P<0.01). In DIR hypothyroid patients, the CRP correlates negatively and significantly with T.Ch (P<0.05) and correlate positively and significantly with F.G and CP (P<0.05). In PIR hypothyroid patients, the CRP correlate positively and significantly with TNF - α (P<0.01). Since the inflammatory marker (IL - 6) increased in insulin resistant cases and known by its critical role in different metabolic reactions in the body, therefore, this study involves a design for an experiment to detect the thyroid gland function and compare its effects with the effect of Carbimazole : An antithyroid drug. The experiment include three groups of rats, each injected with different material : Recombinant human interleukin - 6 (rhIL - 6), Carbimazole , Normal saline (N.S) as control . The results of the experiment show that both IL - 6 and carbimazole cause a decrease in the levels of thyroid hormones (T3,T4) in animal blood , and cause a significant decrease in animal body weight , but there is no effect on the liver weight . There was a significant decrease in glucose level in the animals injected with carbimazole, while in the IL - 6 treated animals there was no change in glucose level. There was a significant increase in the Triglyceride (T.G) and very low density lipoprotein cholesterol (VLDL - C) in the animals treated with IL - 6 , while in the animals treated with carbimazole , there was no change . Finally, injection of the animals with IL - 6 or carbimazole causes significant increase in lymphocyte apoptosis in peripheral blood.

Abstract: تعد امراض الغده الدرقية من الامراض شائعة الحدوث لاسباب مختلفة وتشمل امراض الغدة الدرقية كل من فرط الدرقية (Hyperthyroidism) (وقصور الدرقية ((Hypothyroidism اذ يحدث تضخم الغده الدرقية في الحالتين . يزداد انتاج هرمونات الغدة في مرضى فرط الدرقية بالرغم من انخفاض تركيز الهرمون المحفز للدرقية ( Follicle stimulating hormone )في حين يقل انتاج هرمونات الدرقية في مرضى قصور الدرقية بالرغم من زياده انتاج الهرمون المحفز للدرقية.هنالك علاقة بين هرمونات الغدة الدرقية والايض من خلال تاثير هذه الهرمونات على ايض الكربوهيدرات , الدهون والبروتينات وبالتالي تنتج الطاقة من تحلل هذه المركبات ولقد اجريت هذه الدراسة للتعرف على التاثيرات المختلفة لمعايير الفسلجية والنسيجية والكيموحيوية والجزيئية في مرضى فرط الدرقية ومرضى قصور الدرقية لمجموعتين عمريتين هما اقل من 45 سنه واكثر من 45 سنه للذكور والاناث.اظهرت نتائج الدراسة اختلافات معنوية بين كل من الهرمون المحفز للدرقية وهرمون الدرقية الثلاثي وهرمون الدرقية الثايروكسين وهرمون الايرسين ومستويات مستقبلات الانترليوكين ومستويات الكلكوز بالمصل ومستويات معدل كتلة الجسم ومستويات الكالسيوم بالمصل ومستويات قابليه مضادات الاكسدة بالمصل ومستويات الصوديوم بالمصل في مرضى قصور الدرقية ومرضى فرط الدرقية مقارنه مع مجموعه السيطرة في حين وجدت اختلافات غير معنوية بين انزيم البيروكسيدايزعند مرضى فرط الدرقية في كلا الجنسين ,مستقبل بروتين الثلاثي (FCRL3)والثايروكلوبيولين عند مرضى قصور الدرقية في كلا الجنسين ومستقبل الهرمون المحفز للدرقية في مرضى قصور الدرقية في الذكور والاناث في المرضى الاقل من 45 سنه فقط مقارنه بمجموعة السيطرة ,كما اظهرت الدراسة النسيجية وجود اختلافات في التركيب النسيجي للحويصلات الدرقية بين مرضى الغدة الدرقية بنوعيها (فرط الدرقية وقصور الدرقية) مقارنة مع السيطرة استخدم في هذه الدراسة جين MAPKinaes gene للكشف عن اورام الغدة الدرقية , وقد اظهر استخدام كيمياء المناعة النسيجية وجود ورم سرطاني من النوع الحلمي Papillary Carcinoma في احدى العينات المستحصلة لهذا الغرض .بينت نتائج الارتباط بين المعايير المختلفة لمرضى فرط الدرقية ومرضى قصور الدرقية وجود ارتباط سلبي معنوي p˃0.05)) بين الهرمون المحفز للدرقية معدل كتلة الجسم وارتباط سلبي معنوي p˃0.05)) بين مستقبل الانترليوكين وهرمون الدرقية الثلاثي وارتباط موجبة معنوي p˃0.05)) بين كلا من الايرسين وهرمون الدرقية الثلاثي وعلاقة ايجابية معنوية p˃0.05)) بين كلا من الايرسين وهرمون المحفز للدرقية | The thyroid disease is a common disease of occurrence for different causes which include thyroid hyperthyroidism and hypothyroidism . Goiter occurs in both cases and the production of thyroid hormones increases despite the decline in the concentration of the thyroid stimulating hormone (TSH), while the production of thyroid hormones decreases in hypothyroidism despite the increase in the production of thyroid stimulating hormone. In view of the relationship between thyroid hormones and metabolism and the metabolic rate through the influence of these hormones on the metabolism of carbohydrates, fats and proteins and thus energy produced from the hydrolysis of these compounds This study was conducted to investigate the different effects on different physiological ,histological biochemical and molecular criteria in 58 patients with hyperthyroidism and hypothyroidism for two age groups , they were collected from the Teaching Laboratories in Al - Hilla teaching hospital and from private laboratories (Babylon) , during the period from November 2015 to march 2016, the aim of this study was the detection of the best possible diagnostic parameter for the detection of the dysfunction of thyroid gland by using various biological markers in this study. The results of this study showed that a significant differences in thyroid stimulating hormone (TSH), irisin, thyroxin (T4), triiodothyronine (T3), triglycerides (TG), body mass index (BMI), glucose, Interlukin - 2 Receptor (CD25), total antioxidant capacity (TAC), Potassium (K), Sodium (Na), high density lipoprotein (HDL), calcium (Ca) in hypothyroidism ,hyperthyroidism patients compared with the control group, while the differences were Non - signifecant in the thyroid Peroxidase ( TPO) in patients with hyperthyroidism in both sexes Fc - receptor - like - protein3 (FCRL3), thyroglobulin (Tg) in patients with hypothyroidism in both sexes and thyroid Stimulating Hormone Receptor (TSHR) in patients with hypothyroidism in both sexes only in younger old patients as compared to the control group. Histological study revealed that there were differences in the histological structure of the thyroid vesicles in patients with hyperthyroidism and hypothyroidism compared with the control group. MAP Kinase gene was used for the detection of tumors of the thyroid gland by using immunohistochemistry technique, the result in tissue Papillary Carcinoma was identified in one of the samples obtained for this purpose. The results of the correlation between the different criteria for patients with hyperthyroidism, and hypothyroidism have shown a negative correlation between the thyroid stimulating hormone (TSH) and body mass index (BMI) and interlukin - 2receptor (CD25) and triiodothyronine (T3),while there positive correlation was a between irisin and triiodothyronine (T3) and a positive relationship between both irisin and High Density Lipoprotein (HDL), as well as positive correlation between irisin and thyroid stimulating hormone (TSH).

Abstract: اجريت الدراسة الحالية في الفترة من شهر ايلول2015 ولغاية اب 2016 للتحري عن وبائية الطفيليات المعوية الاميبا الحالة للنسيج Entamoeba histolytica والاميبا غير الممرضة Entamoeba dispar بطريقة الفحص المجهري وطريقة تفاعل سلسلة البلمرة PCR والمقارنة بين كلا الطفيليين في بعض جينات عوامل الضراوة السستين بروتينيز 5 وamoebapor A ، Ap - B ، Ap - C . كذلك تضمنت الدراســـــة تقدير تركـــــيز الضــــد الافرازي S - IgA ، وكذلك قياس تركيز الاضداد IgA ، IgG ، IgMفي مصول المرضى والسيطرة.بلغ عدد العينات المفحوصة 200 عينة خروج لمرضى يعانون من الاسهال لفئات عمرية مختلفة تم جمعها من المستشفيات والمراكز الصحية وبعض العيادات الخاصة في محافظة بابل، تم فحص العينات بطريقة المسحة المباشرة وطريقة تفاعل تسلسل البلمرة باستخراج الدنا الخاص بالطفيليات منها ، في مختبرات المستشفيات والمراكز الصحية ومختبر الطفيليات المتقدم في كلية العلوم/ جامعة بابل .اظهرت نتائج الفحص المجهري ان نسبة الاصابة الاجمالية بالطفيليات المعوية اميبا الزحار 20% ) 40عينة) ، بينما بلغت 73% بطريقة تفاعل تسلسل البلمرة في حين كانت نسبة الاصابة 27.5 % لطفيلي E. dispar باستخدام البرايمر الخاص بهذا الطفيلي Ed - 1. وتضمنت الدراسة الحالية ارتباط عامل العمر والجنس ومنطقة السكن واشهر السنة بنسبة انتشار كلا الطفيليين وباستخدام الفحص المجهري وPCR واوضحت نتائج التحليل الاحصائي عدم وجود فروق معنوية في عدد المصابين بين العمر والجنس الريف والمدينة وباختلاف نوع الطفيلي، بينما وجدت فروق معنوية لنسب انتشار كلا الطفيليين حسب اشهر السنة .بينت نتائج الفحص المجهري حسب الفئة العمرية ان اعلى نسبة اصابة بلغت 37.5% للفئة العمرية (1 - 10) سنة ، بينما بلغت اقل نسبة اصابة 2.5 % للفئة العمرية (31 - 40) سنة، في حين اشارت الدراسة الجزيئية الى ان اعلى نسبة اصابة36.36 و41.3 % للفئة العمرية (1 - 10) سنة واقل نسبة اصابة هي 9.09 و3.4% لكلا من طفيلي E. dispar وE. histolytica ، على التوالي.حسب الجنس بلغت اعلى نسبة اصابة لطفيلي E. histolytica للذكور67.5 % في حين كانت في الاناث32.5 %بطريقة الفحص المجهري ، اي كانت اعلى من نتائج الـــ PCR ، اذ بلغت 41% ، 59 % للاناث والذكور على التوالي ، مقارنة بطفيلي E. dispar 73% للذكور اعلى من الاناث 27%.بلغت نسبة الاصابة بطفيلي اميبا الزحار حسب منطقة السكن في الريف 75% اعل من المدينة 25% بطريقة الفحص المجهري ، مقارنة بنتائج الـــ PCR كانت نسب الاصابة في الريف 69% اعلى من المدينة 31% ، بينما لطفيلي E. dispar في المناطق الريفية 64% اعلى من المدينة 36%.وفقا للاشهر السنوية للدراسة كانت الاصابة اكثر شيوعا في يونيو ويوليو 20و 27.5% على التوالي بطريقة الفحص المجهري، واقل نسب في نوفمبر وديسمبر ويناير ومارس بنسبة 2.5% , مقارنة لنتائج الـــPCR لاميبا النسيج كان الاصابة اكثر شيوعا في يونيو ويوليو 16.6 ، 33.3 % على التوالي، وكان اقل تواترا في اكتوبر، نوفمبر، ديسمبر، ومارس 3.3% ، في حين لطفيليE. dispar كان معدل الاصابة مرتفع في مايو ويوليو 36.36% ، واقل في سبتمبر9.09٪، مع وجود فروقات معنوية بين الطفيليات لكلتا الطريقتين.اوضحت نتائج الدراسة الجزيئية ان هناك تغاير في الدنا الخاص بعوامل الضراوة في كلا الطفيليين ، اذ كانت جميع عينات طفيلي اميبا الزحار موجبة للجين البادئ سستين بروتينيز 5 CP5 وكذلك لــ amoebapor A ، Ap - B، Ap - C، في حين لطفيلي E. dispar وجدت سبعة عينات 63.6% موجبة لــ amoebapore A ، وستة عينات 54.5% لــ Ap - B ، وثلاث عينات 27.2% لــ Ap - C، مع وجود فروق معنوية لكلا الطفيليين . اظهرت نتائـــــــــــــــــج الدراسة المناعية وجود فــــــــــــــروق معنوية للمرضــــــى المصابيــــــن بطفيلي اميبا الزحار مقارنـــــــــــــــة مع مجموعــــــــــة السيطــــــــــرة ، وبلـغ مستوى تركيز الكلوبيوليــــن المناعي S - IgA (8.582 ± 0.351) مايكروغرام/ ديسيلتر لطفيلي E. histolytica و(5.213 ± 0.421) مايكروغرام/ ديسيلتر لــ E. dispar و(1.935±0.03) مايكرو غرام/ ديسيلتر لمجموعة السيطرة ، كذلك بلغ اعلى تركيز لـــــ IgA , IgG, IgM (387.9 , 707.4, 294.9) ملغ/دل لطفيلي اميبا الزحار مقارنة مع طفيلي E. dispar مع مجموعــــــــــة السيطــــــــــرة ، مع وجود فروق معنوية للمرضى المصابين باميبا الزحار . | The present study conducted in the period from September 2015 to Augusts 2016 to determination of epidemiology for Entamoeba histolytica and nonpathogenic Entamoeba dispar by microscopic examination and conventional PCR and compromise some virulence factors cysteine proteinase CP5 gene , amoebapore A , Ap - B, Ap - C, for both parasites and estimate immunoglobulin G,A,M, and S - IgA in serum of patientsAll stool samples (200) collected from patients for different ages suffering from diarrhea or bloody diarrhea and attended to some hospitals and medical centers in Babylon governorate . All samples were examined by direct wet preparation method and polymerase chain reaction by extract the parasites DNA, in hospital and medical centers laboratories and the advanced parasites laboratory in the Biology Department / College of Science / Babylon University. The microscopic examination results show out of 40 stool samples had E. histolytica with percent 20% ,in compromise to PCR product infection rate was higher 73% , and for E. dispar 27.5% by Ed - 1 primer specific for this parasite.This study includes the relation of age, gender, and residence for both parasites infection, without significant differences for both parasites, while the annual study months had significant differences for both parasites by two methods . The microscopic study results according to ages group the highest infection percentage 37.5% for ages (1 - 10) year and the lower percentage was 2.5% for ages (31 - 40) year, while by PCR technique higher percentage 41.3 , 36.36 % for ages (1 - 10) , while the lower percentage was 3.4% , 9.09 % for (21 - 30) years, for E. histolytica and E. dispar , respectively . According to the gender, the infection Percentage in male and female 67.5% , 32.5% respectively by microscopic examination ,is higher than infection rate by PCR product 59% , 41%, for E. histolytica respectively, in compromise for E. dispar in male 73 % is higher than female 27 %.E. histolytica infection according to rural regions was 75 % higher than urban regions 25 % by microscopic examination ,in compromise to PCR product ,infection rate in rural regions was 69 % higher than urban regions 31 %, while for E. dispar in rural regions was 64% higher than urban 36 %.According to annual months of the study, the most frequent infection was in June and July 20 , 27.5% respectively by microscopic examination , and lower frequent was in November , December , January and March 2.5%, in compromise to PCR product results for E. histolytica the most frequent infection was in June and July (16.6 , 33.3)% respectively , and lower frequent was in October, November , December , and March 3.3% , while for E. dispar The high infection rate was in May and July 36.36%, and lower frequent was in September 9.09 %, with significant differences between parasites for both methods. For molecular study results, out of (40) stool samples, only (29) sample showed positive for CP5 gene marker for E. histolytica and 11 (27.5%) samples showed negative for this gene, moreover (11) stool samples showed also positive for (Ed) primer which is specific for E. dispar ,all samples of E. histolytica were positive for cysteine proteinase CP5 gene , amoebapore A , Ap - B, Ap - C, while for E. dispar only seven samples 63.6% positive for amoebapore A, six samples 54.5 % for Ap - B, and three samples 27.2% for Ap - C , with significant differences between both parasites virulence factors genes. The level of secretory IgA showed significant differences in patients with E. histolytica compared with control group, The level of secretory - IgA was (8.582 ± 0.351) μg /dL for E. histolytica and (5.213 ± 0.421) μg /dL for E. dispar and (1.935±0.03) for control group.The result of the present study, revealed the concentrations of immunoglobulin IgA , IgG , IgM, (387.9 , 707.4, 294.9 mg/dl), were higher in E. histolytica patients group compared to E. Dispar patients and control group , with a significant differences at p ≤ 0.05 for E. histolytica group

Abstract: Dexamethasone (DX) is considered one of the steroidal drugs which have varieties of clinical application in medicine for treatment of many different cases. The present study was aimed to know the effect of DX on the implantation of blastocyst during the days 7 and ten of pregnancy, and the process of development of cartilage and bones in the fetuses of albino rats during the days 15 and 20 of pregnancy. In addition to that some physiological parameters aimed to be elucidated in the pregnant rats. The study was conducted in the department of Biology \College of Education for Pure Sciences/ University of Karbala in the period of April, 2016 to May 2017. Ninety four albino rats have been used, among them 10 male rats were used for matting with 84 female rats .The females were divided into three groups, with 28 rats for each group . The first control group was injected with physiological normal saline (0.9%) intraperitoneally. The other two groups were received daily injection via the same route with 0.2 and 0.4 of DX \ 250 gm. of body weight respectively. Each group was further subdivided into four subgroups with seven pregnant female rats for each one. The pregnant rats of each subgroup were sacrificed on the following days : 7,10,15, and 20 respectively The subgroups of the latter two days were used to study the development of the cartilage and bones. Half of the embryos on day 20 of pregnancy from the control and treated groups were used for histological sectioning; while the other half were used to stain the embryonic skeleton according to double staining technique of Whitaker and Dix . All the collected samples which include the implantation sites from the pregnant uteri at days 7 and 10, together with whole embryos from days 15 and 20 of pregnancy, were put in two types of fixatives : 10% formalin and Boun's solution. The collected samples then processed according to the traditional methods of paraffin embedding to be cut into sections of five µm thicknesses. The sections were stained by haemotoxylin and eosin (H&E); one step trichrome; periodic acid Schiff's (PAS) with and without diastase; Alcian blue at pH 1.0 and 2.5. Blood samples from pregnant rats were collected on days 7,10,15,and 20 of pregnancy to check for the following biochemical parameters : concentration of calcium ,phosphate and potassium ions ;alkaline phosphatase ,and acid phosphatase enzymes; and estimation of parathyroid and calcitonin hormones. The results of the H&E stained section of pregnant sites of both the control and treated groups have shown that blastocyst was implanted in the antimesometrial side of the uterus. Most of this side of the uterus has become decidualized tissue, where four zones could be identified : the primary decidual zone; secondary decidual zone ;implantation site; and undifferentiated decidual basal zone. On day 10 of pregnancy the embryos have shifted their position from the antimesometrial side to the mesometrial side of the uterus. The decidual tissue has grown wider and included most of stromal cells of the endometrium. The changing of the position of the embryos at day 10 is important for their survival because they will become nearer to the main blood supply to uterus which are running in the mesometrium. The results of the one step trichrome stained sections from both the control and treated groups have shown clear distribution of collagen fibers in the decidual tissues at days 7 and 10 of pregnancy, which gave an indication of the success of pregnancy up to these stages. The primary decidual zone was devoid of collagen fibers but they are evident in the secondary decidual zone. At day 10 of pregnancy the collagen fibers were more in evidence especially at the mesometrial side of the uterus. The importance of collagen fibers was to give support to the blood vessels supplying the placenta which are coursing through the mesometrium. Positive stain has been revealed in PAS reaction in decidual tissue on day seven of pregnancy without diastase, but it was negative with diastase. No reaction ,in the decidual tissue, with and without diastase was noticed at day ten of pregnancy. With Alcian blue pH 1.0 no reaction was noticed in the decidual tissue of both control and treated groups at days seven and ten of pregnancy. On the other hand positive reaction has come out at pH 2.5. The reaction at that pH was more prominent at the antimesometrial decidual tissue. But it was only depicted in the wall of blood sinusoid of the mesometrial decidual tissue. Tongue like extensions of trophoblasts was noticed to be extending from the recently implanted embryo toward the endometrium, at day seven of pregnancy of both the control and treated group. The trophoblasts were pushing themselves in the direction of mesometrial decidual tissue in order to reach and settle nearer and in the wall of the blood sinusoids of that area. The invasion trophoblasts destination was to reach to the spiral arteries supplying the labyrinthine placenta in the rats. At the 15th day of gestation there was no significant change in the cartilage model in the posterior limbs of the two treatment groups from the control group. The important signal indicated by the present study was observation of cartilaginous cell necrosis, hyperplasia and inflammatory cell infiltration, At the age of near the end of pregnancy in the treatment groups, many of the effects on the bone tissue, which are the existence of a vacuum around the bone cells and the infiltration of inflammatory cells in addition to bone erosion and arthritis and the reduction of cartilage and irregularity of medicine Surface area, hyperlipidemias and leavicular necrosis. Bone marrow squeaks in the control group and the first dose group of dexamethasone were found to be more extensive than the second treatment group. The bones of control group embryos stained with alizarin red has acquired much more red coloration than the treated group embryos, which indicated a slowdown of ossification in their bones compared with control group, Showed that the length of the skull, spine, forelimbs and hind limbs of the fetal fetus at the age of 20 days of gestation of from the treatment groups has decreased for length in the control group but the decline was not significantly level (P <0.05) . The calcium, phosphate and potassium ions concentration was low in the blood serum of treated group ( P< 0.05) during the whole period studied. However the phosphate ions was low in the control group .For the alkaline phosphatase level, it was significantly low ( P< 0.05) in the treated groups compare with the control. On the other hand the level of the acid phosphatase was high in the treated group ( P< 0.05). The concentration of both the parathyroid and calcitonin hormones was significantly higher ( P< 0.05) in the treated groups compared with control. In conclusion, the present study has revealed that DX treatment showed no interference with the process of implantation during the days seven and ten, and with chondrogenesis on day fifteen of pregnancy. While the effect on cartilage formation on the fifteenth day and bone on the twentieth day of pregnancy.

Abstract: The objectives of current study were to identify Trichoderma viridi and Trichoderma harzianum by genetics methods and used them singly or both (3% for each treatment) in improvement of barley straw and animal performance. A total of 20Arabi lambs aged about 3 - 4months with an average weight of 22.725±1.43 kg distributed randomly and equally to four nutritional treatments. Lambs fed individually as 2% of body weight, while straw was given ad libtium. The results obtained can be summarized as follows : 1 - The study was able to identify TV and TH using three different PCR techniques, which are PCR - RAPD, PCR - ITS and PCR - IGS.TV and TH showed three and two bands respectively when using PCR - RAPD at nearly 600bp.While both fungi showed one band by using PCR - ITS and PCR - IGS and at nearly 600 and 2,400bp respectively.2 - Using both fungi (treatment 4) resulted in a significant improvement in the chemical composition of the straw through the high content of crude protein and low crude fiber, ADF and NDF compared to untreated straw.3 - There were improvements indigestibilityofcrude protein, fiber, cellulose, hemicellulose, ADFand NDF when the both fungi treatment was used. As well as total digestible nutrients of the fungus treated groups in comparison with untreated straw (control group).4 - Feed intake of treated straw groups increased significantly compared to untreated straw groupwiththe superiority of the group that was treated with both fungi (639.91 - 826.30 gm/head/day for control and both fungi group respectively). As well as there wasa significant improvement in feed conversion efficiency of a group treated with both fungi in comparison of other groups (6.38 - 5.52 kg feed/kg growth rate respectively).5 - Rumen pH, ammonia and total volatile fatty acid of lambs fed treated straw (6.22 - 6.89, 10.00 - 18.69; 11.35 - 13.61 for control115group and both fungi group respectively) were significantly higher than those of untreated group.6 - There were significant improvements in lambs’bodyweigh tsand their absolute and relative gains when fed treated straw (37.82 kg, 14.97kg and 65.67% respectively) especially the group fed both fungi in comparison with untreated group (32.79 kg, 10.03kg and 43.89%respectively).7 - Globulin, AST and ALT was not significantly influenced bytreated straw. On the contrary, there were a significant increase inhemoglobin, glucose, total protein, albumin and albumin/globulin ratio of the treated groupcompared with the untreated straw. While a significant decrease in urea and cholesterollevelin theblood serumof treated groups in comparison with untreated group.8 - Economic evaluation ofadding fungitostrawshowed improvement inthe relativeeconomic efficiencyby26.2%, 37.0% and 59.5% for adding TVorTH or bothtogetherrespectively. Adding fungi decreasedthe cost of feed from 46.65Dinars/ kg increase in body weight gains incase of untreated straw to 40.15dinars/ kg when adding both fungi.

Abstract: This study was undertaken at the poultry field of Animal ResourceDepartment \college of Agriculture \University of Basra in Jan.,28th toMar.,3th,2013 and in Oct.,28th to December,23th 2013 for the first and secondexperimentrespectively . The objective of this study was to evaluate the effectof hatching eggs injection with the some nutritious (Folic acid vitamins,Pyridoxine, Threonine amino acid) for the first experiment and choosing besttreatment from first experiment and injection with Arginine amino acid ,folicacid as well as the effect of feeding time on the performance and somephysiological traits of broiler chicks .In the first experiment ,825 hatching eggsof Ross 308 broiler were received from the hatchery .At 14th day of incubationstage ,eggs were tested and unfertile eggs were discarded ,the remainder eggswere divided into seven treatments and injected with 0.1 ml of solution in airsac position of the egg as follows : 1. First treatment : injection 100 eggs with 0.1ml of threonine solution (30μgconcentration per an egg).2. Second treatment : injection 100 eggs with 0.1 ml of b6 vitamin solution(100μg concentration per an egg)3. Thirdtreatment : injection 100 eggs with 0.1 ml of mix from L - threonine aminoacid plus b6 vitamin (30μgand 100μg) per an egg.4. Fourthtreatment : injection 100 eggs with 0.1 ml of Folic acid vitamin solution(250g) per of an egg.5. Fifthtreatment : injection 100 eggs with 0.1 ml of mix solution from Lthreonineamino acid plus Folic acid vitamin (30μg and 250 g) per an egg.6. Sixthtreatment : positiveControl : injection100 eggs with 0.1 mlSterile distilledwaterper egg.7. Seventhtreatment : negativeControl : 155 eggs were left withoutinjection.Hatching chicks of each treatment were distributed in to three replicate (15 Chicksfor each replicate) .Birds were reared up to five weeks of age.The results of first experiment painted out to : 1. There was significantly (P<0.05) proceeding for the treatment of egg injectionwith some nutrients on hatching percentage ,hatching chick weight ,bodyBweight ,body weight gain and weekly and Cumulative feed conversion ratio.whereas, this treatment showed less embryonic mortality percentage incomparison with other experimental treatments and There was significantly(P<0.05) increase in weekly and Cumulative feed intake of birds that werehatched from injected eggs with ( folic acid ,threonine amino acid) during thesecond ,fourth and fifth week were as ,the treatment of pyridoxine injectionshowed less mortality percentage compared to other experimental treatments .2. Treatment of folic acid and threonine and pyridoxine injection showedsignificant (P<0.05) increases in the dressingpercentage, weights of breast,thigh and edible viscera comparison with other treatments.3. There was significantly(P<0.05) increase in red blood cells number,hemoglobinconcentration package cells volume of broilers that were hatchedfrom egg injection with (folic acid plus threonine) at 3 and 5 weeks of agecompared to other treatment While , Treatment of Pyridoxine eggs injectionshowed highest mean of white blood cells at 3 weeks of age, whereas,treatment of Pyridoxine and folic acid eggs injection proceeding otherexperimental treatment in highest mean of white blood cells at 5 weeks of age .4. There was significantly(P<0.05) increase in total protein concentration,glucose ,Globulins, and less uric acid concentration and cholesterol in chicksblood serum of folic acid and threonine egg injection treatment at 3 and 5weeks of age in comparison with other treatment. Whereas, there were nosignificant effect of egg injection on triglycerides concentration and on T3 andT4 hormone in broiler blood serum.5. There was no significantly effect of egg injection with nutritive on low densityfatty proteins concentration whereas,the negativecontrol treatment showedhighest concentration of low and high proteins at 3 and 5 weeks of agecompared to other experimental treatments.6. T4 hormone concentration in hatchingbroiler blood serum of threonineinjection treatment increased significant (P<0.05) at 3 weeks of age ,whereas,T4 hormones concentration was higher respecting treatment of folic acid andthreonine injection at 5 weeks of age in comparison with other treatment.7. There was significantly (P<0.05) increase in phosphorus ,calcium and ferricconcentrations in blood serum of the hatchingbroilers of folic acid andthreonine egg injection treatment at 3 and 5 weeks of age in comparison withother treatments.C8. There was significantly (P<0.05) increase in the villus lengths cryptsdepth ofjejunum of the boilers Caracasrespecting Folicacid and Threonine injectiontreatment in comparison with other treatment.According to the firstexperiment results and choosing the best treatment of egginjection with nutritive, the second experiment was carried out .the treatmentof this experiment whereas follows : 1. Firsttreatment : hatching eggs injection with (30μg of threonine, 250g of folicacid) per an egg.2. Secondtreatment : hatching eggs injection with (5 μg of arginine) per an egg.3. Thirdtreatment : hatching eggs injection with (5 μg of arginine plus 250 goffolic acid) per an egg.4. Fourth treatment : the control treatment (withoutinjection).Hatching chicks of each treatment were divided in to groups each of threereplicates ( 12 Chicks for each replicates ) .firstgroups chicks were fed at thehatchery ,whereas,second groupschicks were fed after 24 hours at field. Chickswere reared from 1 day after hatching up to 5 weeks of age .the results of thesecond experiment pointed out to : 1. Treatments of injection with arginine and Folic acid and threonine plus folicacid proceeding significantly (P<0.05) in hatching percentage andhatchingchicksweight, whereas, This treatment showed less percentage of lateembryonic mortality in comparison with other treatments. Also, Broiler birdsof Arginine and Folic acid injection treatment showed significantly (P˂0.05)increase in body weight, weekly body again, weekly and Cumulativefeedintake,weekly and Cumulative feed conversion ratio and dressing percentage.Whereas, treatment with arginine and folic acid injection showed less totalmortality mean compared to other treatments. Also, there was significantly(P˂0.05) effect of feeding time on some productive traits. Treatments offeeding at the hatchery showed significant increase in body weight, bodyweight gain, feed intake, better feed conversion ratio and dressing percentagein comparison with treatments of feeding at field after 24 hours.2 - There was a significantly (P˂0.05) effect of interaction between eggs injectionwith nutritious and feeding time on body weight, body weight gain and feedconversion ratio. Whereas,treatment of arginine and folic acid injection showed a significantly (P˂0.05)increase in breast fullness liver and heart weight in comparison with othertreatments.4 - Feeding time affected significantly (P˂0.05) on some carcasses traits. Carcassesof feeding at hatchery treatment were better significantly in body fullness,breastfullness, and heart, liver, gizzard, and Fabricia gland and pancreasweights. Whereas, carcasses of feeding at field treatment were of high pad fatweight. There was no significant effect of interaction between injection andfeeding time on carcasses traits.5 - There was a significant (P˂0.05) increase in red and white blood cells numbers 3days and 5 weeks of age and in hemoglobin concentration at 5 weeks of agerespecting the treatment of arginine and folic acid in comparison with othertreatments. Control treatment exceeded in Heterophil cells\ lymphocyte cellspercentage at 3 and 5 weeks of age compared to treatments.6 - There was a significantly (P˂0.05) effect of feeding time on some cellular bloodcharacteristics. Treatment of the field feeding exceeded in red blood cellsnumbers, hemoglobin concentration and Heterophil cells\lymphocyte cellspercentage at 3 days and 5 weeks of age. Whereas, treatment of the hatcheryfeeding exceeded in white blood cells numbers compared to treatment of thefield feeding after 24 hours.There was no significantly (P˂0.05) effect ofinteraction between injection and feeding time on the cellular bloodcharacteristics at 3days of age, whereas this interaction affected significantly(P˂0.05) on the white blood numbers and Heterophil cells\lymphocyte cellspercentage at 5weeks of age.E7 - Treatment of arginine and folic acid injection exceeded significantly (P˂0.05)in serum total protein concentration at 3 days and in total proteinconcentration, uric acid and GPT, GOT enzymes in blood plasma at 5 weeksof age. Treatment of the hatchery feeding exceeded in glucoseconcentration and total protein at 3 days of age and in total protein concentration and uric acid at 5 weeks of age. There was no significant (P˂0.05) effect ofinteraction between injection and feeding time on the biochemical bloodcharacteristics at 5 weeks of age. 9 - There was a significant (P˂0.05) increase in villus Length and crypts Depth of the intestinal jejunum of comparison with of threonine and folic acid injection treatment in comparison with other treatments. Treatment of the hatchery feeding exceeded (P˂0.05) respecting those traits compared to the field eeding treatment. There was a significantly (P˂0.05) effect of interaction (TheHatchery Feeding) and feeding time villus, length and crypts Depths werehighest at 1 and 5 weeks of age.

Abstract: This study was conducted in the Animal Farm of the College of Agriculture - University of Basrah. A total of 28 Arabi male lambs were used, with average age of 5 months and mean body weight of 21.74 ± 1.33 Kg. Lambs were distributed equally and randomly into 7 nutrition rations in two experiments. First experiment was a growth experiment which lasted for 90 days. The second was digestion field experiment and lasted for 13 days after the end of the first experiment. The experimental rations were as follows : Ration - 1 - (control) - fed on concentrate ration only by 3% of body weight. Ration - 2 - fed on 90% of the control ration and 10% of ground date stones substituted for the barley.Ration - 3 - lambs fed on the ration 2 with addition of 3 grams of bakery yeast (Saccharomyces cerevisiae) / Kg of feed. Ration - 4 - fed on 80% of the control ration with substitution of 20% of date stones for barley.Ration - 5 - lambs fed on the ration 4 with addition of 3 grams of bakery yeast per Kg of feed.Ration - 6 - fed on 70% of the control ration and 30% of date stones.Ration - 7 - lambs fed on the ration 6 with addition of 3 grams of bakery yeast per Kg of feed.Results were as follows : 1 - Mean final body weight (34.00 Kg) except Control ration, total gain weight (12.75 Kg) except the control and 2 rations and daily gain weight (141.67 g/ day) of lambs in ration 3 were significantly higher than animals in other rations.2 - Animals of ration 3 recorded the highest feed consumption arithmetically (0.829 Kg/ day/ lamb) and the best feed conversion ratio arithmetically too (5.85 Kg dry matter/ Kg body weight gain).3 - Substitution of date stones with addition of bakery yeast affected digestion coefficients of feed nutrients. Ration 3 was superior in digestion coefficients of dry matter, crude protein and nitrogen free extract to other rations. These values were 70.64, 72.11 and 80.64 %.4 - Results showed significant differences between means of pH of rumen fluid and total bacterial count which Significant decreases in each of 6 and 7 rations than other rations while in the counts of cellulolytic digestion bacteria, ration 3 were significantly than animals in other rations but not differ significantly from that of the ration 5 while it was similar with control ration. 5 - No significant differences between rations were noticed in means of red blood corpuscles, white blood cells, hemoglobin, packed cell volume, cholesterol concentration, glucose concentration and concentration of the enzymes GPT and GOT. While differences between means of blood urea concentration and total protein of blood were significant.6 - Mean empty body weight, hot carcass weight, dressing percentage on the basis of live weight and empty body weight of lambs in ration 3 28.02 Kg, 16.18 Kg, 47.59% and 57.74% respectively were significantly higher than that of animals in other rations except control ration in hot carcass weight.7 - The highest weights of rib cut (rib 7 - 12), leg cut and loin cut were for animals in ration 3 (1.61, 5.00 and 2.40 Kg respectively) than other rations but in the loin cut, the ration 3 was similar to the control ration 2.16 Kg.8 - For body measurements, the highest leg circumference were recorded for animals in rations 2 and 3 (35.00 and 36.50 cm respectively) than other rations but these rations were similar to the control ration. While the highest rib eye muscle area was for carcasses of animals in ration 3 (14.41 cm2) but this value did not differ significantly from that of the control animals (13.45 cm2).9 - In the case of some carcass muscle weights, the muscle Supraspinatus weight were higher in the animals of the ration 3 (176.67 g) but it was similar to control ration (164.25 g). Weight of the muscle Biceps femoris was higher in the carcasses of animals in ration 3 (625.75 g).10 - Carcasses of animals in ration 3 recorded the highest dissected lean meat 931.70 g and fat 280.55 g in rib cut, but no significant differences were registered for dissected bone weight among the different rations. Also weights of the internal organs of the animals namely : liver, heart, kidneys, lungs, spleen and the testicles for animals in ration 3 were higher than that of other rations.11 - Mean weights of separated fat of carcasses were higher for animals of the 2 and 3 rations (1.93 and 1.95 Kg respectively) in comparison with animals of other rations but these rations were similar to the control ration 2.08 Kg.12 - Substitution of date stones in percentages of 10% and 20%, with the addition of bakery yeast caused significant improvement in organoleptic characteristics (color, tenderness, juiciness and overall acceptability). Evaluation of samples of rations 3 and 5 were superior to that of other rations.13 - Rations 3 and 5 gave the best results for water holding capacity in meat. No significant differences were obtained in cholesterol concentration and pH of meat.Results showed that the lowest values of cooking loss were in ration 3 (43.30 %).No significant differences were observed in chemical composition of theLongissimus dorsi muscle (moisture, protein, fat and ash) between rations.

Abstract: The present study was conducted at the agricultural experimental Research Station / College of Agriculture / University of Basrah form 3rd /1/2015 to 3rd /1/2016. A total of 12 mature rams (4 Najdi , 4 Arabi and 4 crosses) aged 3 - 3.5 years with nearly similar weights . Nutrition depended on concentrate ration ( 47 % wheat bran , 50 % barely ,1% urea , 1% CaCo3 and 1% Nacl ) , with 16.58 % crude protein and TDN 72.17% and was given 0.5kg /animal twice aday (at morning and afternoon)500 - 750 g / animal green fodder with wheat straw was given as it was available in the farm , cubic mineral and water provided freely at the day. Rams were training for semen collection for 30 days before the beginning of the study by using artificial vagina used for sheep. Semen was collected three time each month. The study includes two experiments : First experiment : It was designed to study the effect of breed , season ( spring , summer , autumn and winter ) and month on semen characteristics physical , sexual hormones ( testosterone , ICSH and SSH) of blood serum and level of enzymes ( AST , ALT and ALP ) of blood plasma.Second experiment : It included ten diluters from local materials (Beet pulp juice ) with 5 , 10 and 15% egg yolk and amino acids ( Glycin and cysteine ) with concentration of (5, 10 and 15) mmol. It also included the effect of type of dilutor , cooling , freezing storage period ( one and two months) in liquid nitrogen ( - 1960 C) on individual motility and live sperm percent .bThe results can be summarized as follow : 1 - Arabi rams exceeded (P<0.05) Najdi and crosses rams in their semen pH , spring also showed higher (P<0.05)pH followed by winter in comparison with summer and autumn .May showed higher (P<0.05) semen pH in comparison with other months.2 - Najdi rams exceeded (P<0.05)Arabi and their crosses in mean ejaculate volume, mass and individual sperm motility , sperm concentration , live sperm percent and mean testes circumference .3 - Summer and autumn showed higher (P<0.05) in meanejaculate volume , mass and individual sperm motility , sperm concentration , live sperm percent and mean scrotal circumference than winter and spring . Spring also showed higher (P<0.05) dead sperm percent followed by winter in comparison with summer and autumn .4 - August showed higher (P<0.05) ejaculate volume and mass and individual sperm motility followed July and June in comparison with other months. September , August and October exceeded (P<0.05)in comparison with other months sperm percent . October showed higher (P<0.05) in live sperm percent in comparison with other months . Highest (P<0.05) dead sperm percent happened during March followed by April and May . Scrotal circumference was high (P<0.05) during September followed by November and October.5 - Najdi and Arabi rams showed significant (P<0.05)differences in testosterone and ICSH hormones in comparison with their crosses . However , crosses exceeded (P<0.05)Arabi and Najdi rams in mean of SSH hormone. Autumn followed by summer exceeded (P<0.05) wintercand spring in testosterone , ICSH and SSH hormones. September and October showed higher (P<0.05)testosterone, ICSH and SSH hormones in comparison with other months.6 - Arabi rams exceed (P<0.05) both Najdi and their cross by enzymes concentration ( AST, ALT and ALP) of semen plasma . Spring and winter exceeded (P<0.05) summer and autumn by mean of ALT and AST enzymes. However , summer and winter exceeded (P<0.05) autumn and spring by mean of ALP concentration . March showed highest (P<0.05)AST and ALT concentration , while August showed highest (P<0.05) ALP concentration.7 - Amino acid diluter ( 15 mmol glysin and cysteine ) and 10 % beet pulp juice showed highest (P<0.05) individual motility after cooling and freezing for month and two monthsduring all months of year. While the beet pulp juice 15% and control showed the lowest means (P<0.05) of individual motility after cooling and two months of freezing.8 - Glysin 10 and 15 mmol , cysteine 10 mmol and beet juice 10% showed significant (P<0.05) increase in live sperm percent after and cooling and freezing . On other hand beet pulp juice 15% and control recorded the least value (P<0.05) . Beet pulp juice and control showed the highest (P<0.05) level of dead sperm percent after cooling and freezing in contrary with glysin 10 mmol and cysteine 15 mmol

Abstract: This study was conducted in both Poultry Farm/Animal Resources Department/ College of Agriculture / University of Basra and laboratories of Molecular Biology/ London/ United Kingdom (The Molecular Oncology Unit Laboratories of Guy's and St Thomas's Hospital/ Kings College London/ UK). Molecular analysis of the samples and the detection of a number of gene expression of several feed transporter genes in the small intestine of broiler chickens when fed diets with different levels of energy and protein. A total of 225 unsexed one day old broiler chicks (Ross breed) were randomly distributed on five experimental treatments each with three replicates of 15 chicks. Feeding was ad libitum during the whole period of experiment that lasted 35 days.The first treatment was control with 22.3 and 20 % crude protein and 2910 and 3001 kcal/kg for starter and growth period respectively. The second treatment consisted 24.1 and 22 % crude protein and 2920 and 2978 kcal/kg for starter and growth period respectively. The third treatment consisted 20 and 18 % and 2898 and 2996 kcal/ kg for starter and growth period respectively. The fourth treatment consisted 21.9 and 20 % and 3107 and 3199 kcal/ kg for starter and growth period respectively. The fifth treatment consisted 22.2 and 20 % and 2720 and 2800 kcal/ kg for starter and growth period respectively. The results revealed the followings : - The study showed the ability to amplify all genes and find their abundance according to their primers. - The highest value of APN gene abundance of female in the duodenum (1.86 ng) fed diet with 22 % cp and 3199 kcal/kg ME. - Highest abundance (6.19 ng) of Pept1 mRNA showed by males in their jejunum, when fed a diet consisted 22 % cp and 3199 kcal/kg ME. - The greatest value of b - AT and CAT2 amino acids transporter genes effect was revealed by males fed diet with 22 % cp and 3001 kcal/kg ME. - Females fed 20 % cp and 3199 kcal/kg ME diets reordered highest SGLT1 gene expression (4.70ng) within their ileum. - Males fed 20 % cp and 3199 kcal/kg ME diets reordered highest GLUT5 gene expression (6.37 ng) within their ileum. - All genes associated positively and significantly with body weight, growth rate, carcass cuts percentages (legs, breast, intestine length and weight) at the age of 35 days and negatively with feed consumption and feed conversion ratio. - Amino acid and peptides transporter genes abundance improved with an increase in diet protein, that of saccharide transporter improves with the increase of ME level in the diets. - Male gave higher mRNA copies of most genes in comparison with females, except that of SGLT1 and NPA which increased in females. - It is possible to manipulate nutrient transporter gene expression by feeding. That may reflect on birds performance.

To achieve all the study requirements, the laboratories work at the Laboratory physiology, Molecular Genetics Laboratory, College of Agriculture, University of Basrah; the Central Research Unit, College of Veterinary Medicine, University of Basrah; the Criminal Laboratory Department of Investigation, Criminal Evidence Directorate, Basrah Governorate Police; as well as laboratories of the Abu Ghraib, Artificial Insemination Center; in addition to the First BASE Laboratories, Laboratory in Selangor Malaysia, were used. The study included two experiments : The first experiment : This experiment was designed to detect the genetic polymorphism of hsp70 gene in the semen of Bulls, their relationship to certain physical and chemical semen characteristics, the level of gene expression in the season winter and summer, and the evaluation of the bulls by finding the breeding value of certain traits in the fresh and frozen semen of the Iraqi Holstein Bulls, back to the Artificial Insemination Center / Abu Ghraib / Baghdad, 30 bulls were used in this experiment.Second Experience : This experiment was designed to detect the genetic polymorphism of hsp70 genes in the semen of Rams, their relationship with some physical and chemical semen characteristics, the level of gene expression in the season winter and summer, and the evaluation of the rams by finding the breeding value of some traits in the fresh and frozen semen of local Arabi rams, back to the Agricultural Research Station / College of Agriculture / University of Basrah, 20 Rams were used in this experiment.Results of the study : I. The first experiment : 1) After DNA extracted, the quantity and purity of the DNA were determined by the Nano drop device, the purity ratio was 1.8. A Polymerase Chain Reaction (PCR) carried out, the amplification product was approximately 1926bp. Nucleotide Sequence analysis was done to detect polymorphism and compared with the genotypes of the hsp70 gene in the Gene Bank. There are three Genotypes of hsp70 gene were obtained in bulls : : The Genotype A, this genotype is the closest to heat shock protein hsp70 gene in the Holstein bulls in Gene Bank. This polymorphism was exposed to be genetic mutations at position No. 6, the Nucleotide (C < G), No. 174 (C < G), No. 282 (A< G) and position No. 1339 (T < C). All mutations are silent that did not encodingto any new amino acid, (16 Bulls).B : The Genotype B, the nucleotides were substituted at different positions, position No.114 (G < A), No. 174 (C < G), No. 282 (A < G), No. 1339 (T < C),No.1451 (C < A), No.1590 (A < G), No. 1695 (C < T) and position No. 1719 (G<T), these genetic mutations are all silent except the mutation at the position No.1451 is a missense mutation, encoding for new amino acid, (6 Bulls).C : The Genotype C , the nucleotides were substituted at different positions, position No. 114 (G < A), No. 174 (C < G), No. 282 (A < G) No.1339 (T < C),No. 1590 (A <G), No. 1695 (C <T) and position No. 1719 (G <T), all mutationsare silent, ( 8 bulls).2) The results of the Multiple Sequence Alignment (MSA) of hsp70 gene in the bulls showed that the convergence ratio of A, B and C were 99.73%, 99.51% and 99.56%, respectively, with the same gene in the Holstein bulls recorded in Gene Bank. The MSA results also showed that the B genotype gave the lowest ratio to the studied genotypes, followed by the C genotype and the A genotype, which showed the highest approximation with the genotypes studied in the Gene Bank for the Holstein bulls, sheep, goats, pigs and humans.3) Genotype B, has shown significantly higher (P < 0.05) than the C genotype and A genotype in all physical semen characteristics studied, it gave the highest rate of mass and individual motility, the highest percentage of live sperm and thelowest rate of abnormalities, both in fresh and frozen semen in a period of onemonth and two months freezing and in both seasons of the study.4) Genotype B showed Significantly higher (P < 0.05) towards the C and A genotypes in all chemical semen characteristics studied, It gave the highest percentage of the Acrosome integrities and gave the lowest level of concentration of enzymes ALT, AST and ALP in the frozen semen and freezing periods in a month and two months and in both seasons of the study.5 ) After RNA extraction, synthesise of cDNA and carry out Quantitative Realtime PCR (qRT - PCR) analysis, genotype B showed Significantly higher (P < 0.05) than genotype C and A in the level of gene expression of heat shock protein hsp70 gene in the summer season.) The bulls belong to genotype B were superior in the valuation on the basis of the breeding value of motility, live sperm and the abnormalities in the fresh and frozen semen for a month and two months followed by the C than A.II. Second Experience : 1) After DNA extracted, the quantity and purity of the DNA were determined by the Nano drop device, the purity ratio was 1.8. A PCR carried out and the amplification product was in approximately 1926bp, bands were visualized by gel electrophoresis. Nucleotide sequence analysis was done to detect polymorphism compared to that of the hsp70 gene in the Gene Bank. There are two genotypes ofhsp70 gene were obtained in rams : A : The A genotype, showed no match with other nucleotides sequence of other group neither heat shock protein hsp70 gene in Gene Bank, the nucleotide in position 1308, the nucleotide T was substituted by C (T < C), a silent genetic mutation that did not encoding a new amino acid, (10 rams).B : The B genotype, in comparison with the A genotype , the Nucleotide C was substituted with G (C <G), a missense genetic mutation that resulted from change the amino acid encoding at the site 514, a genetic mutation not previously recorded in rams except in inese sheep. In addition, nucleotide G was substituted by nucleotide A (G < A) in the position 1524, a silent genetic mutation (10 rams).2) The results of the (MSA) of hsp70 gene in the rams showed that the ratio of Aand B genotypes were 99.89% and 99.84% respectively with the same gene inrams in the Gene Bank. The MSA of the genotype B gave the lowest ratio to thesame gene in sheep, Holstein bulls, goat, pig and human3) The genotype B Significantly higher (P <0.05) than the genotype A in allPhysical semen characteristics studied, it showed the highest rate of mass andindividual motility and the highest percentage of live sperm and the lowest rate ofabnormalities, in fresh and frozen semen in a period of a month and two monthsfreezing and in both seasons of the study.4) The genotype B Significantly higher (P <0.05) than the genotype A inall chemical semen characteristics, it showed the highest percentage of theAcrosome integrities, and showed the lowest level of concentration of enzymesALT, AST and ALP in the frozen semen and freezing periods of month and twomonths and in both seasons of the study.

The aim of this study was to investigate the effect of early (8 - 21) and late (21 - 34) day of feed restriction by partial 20% with adding IRAQI probiotic at level 0.3% for (first experiment) and propionic acid at level 0.3% for the (second experiment) in productive performance and some physiological of blood and small intestine. One day of 270 unsexed chicks of Ross 308 strain were used in the study. The chicks were randomly distributed into 6 treatments with three replicates. Each replicate contain 15 chicks. Results of first experiment indicated that : 1 - There was a significant (p<0.05) increase in control treatment with IRAQI probiotic on live body weight and body weight gain. While, there was a significant (p<0.05) decrease in the late feed restriction treatment with, or not adding probiotic.2 - There was a significant (p<0.05) increase for the control treatment with, or not adding probiotic in feed consumption.3 - There was a significant (p<0.05) improvement in feed conversion ratio of early and late feed restriction with, or not adding probiotic.4 - Late feed restriction and early feed restriction with probiotic cause significant (p<0.05) decrease in mortality comparing with control treatment.]b[5 - There was a significant (p<0.05) decrease in abdominal fat for the treatments early and late feed restriction with, or not addition probiotic as compared with the control treatment.6 - There was significant (p<0.05) reduction in Intestine PH for the treatment adding probiotic, but there was no differences between the treatments in Ceca PH.7 - There was a significant (p<0.05) decrease in total protein, albumin, cholesterol and glucose in blood serum for the chicken with early feed restriction (not adding probiotic), and there was significant (p<0.05) increase GOT and GPT levels in the same treatment at the age of 21 days, Addition of IRAQI prpbiotic showed a significant (p<0.05) reduction of serum cholesterol, GOT and GPT as compared with control treatment.8 - There was a significant (p<0.05) decrease in cholesterol, GOT and GPT by the addition IRAQI probiotic compared with control treatment at 42 days.Results of second experiment indicated that : 1 - There was a significant (p<0.05) decrease in the treatments (late feed restriction, early and late feed restriction with, or not adding propionic acid) on live body weight and body weight gain.2 - There was significant (p<0.05) decrease in feed consumption for the treatment early and late feed restriction with, or not addition propionic acid. 3 - There was significant (p<0.05) decrease in feed conversion for the treatment early and late feed restriction with addition propionic acid compared with the other treatments.4 - Late, early feed restriction with or not addition propionic acid cause significant (p<0.05) decrease in mortality compared with control treatment, and there was significant (p<0.05) reduction in production index for the treatment early and late feed restriction with addition propionic acid as compared with the other treatments.5 - There was a significant (p<0.05) decrease in abdominal fat for the treatments early and late feed restriction with, or not addition propionic acid compared with the control treatments.6 - There was a significant (p<0.05) decrease in total protein, globulin, cholesterol and glucose in blood serum for early feed restriction with, or not adding propionic acid, but there was significant (p<0.05) increase at the level of GOT and GPT in the same treatment compared with the other treatments at the age of 21 days.7 - Serum cholesterol showed significant (p<0.05) reduction in the late feed restriction with, or not adding propionic acid compared with the other treatments at the age of 42 days.

Abstract: اجريت هذه الدراسة في محطة البحوث الزراعية التابعة الى كلية الزراعة بجامعة البصرة وبعض الحقول الاهلية المجاورة لجامعة البصرة في كرمة علي واستخدم في الدراسة 23 بقرة مضربة باعمار ومواسم مختلفة حيث اعتمدت التغذية الحرة على النخالة والقصب واجريت القياسات للفترة من 1/12/2006 - 31/12/2007 لدراسة تاثير مقاييس واشكال الجسم والضرع والحلمة على انتاج الحليب ومكوناته اليومي والكلي والجزئي والمعدل لـ305 يوم ودراسة منحنى انتاج الحليب ومكوناته .واظهرت النتائج مايلي : 1 - بلغ متوسط انتاج الحليب اليومي والكلي والمعدل لـ305 يوم 5.80 و1740.97 و1883.91 كغم على التوالي وتاثر كل منهم معنويا (p<0.05) بكل من مرحلة وموسم الادرار, وتاثر انتاج الحليب المعدل لـ305 يوم معنويا (p<0.05) في كل من شكل الضرع وشكل فتحة الحلمة وشكل الحلمة , بينما لم يكن لشكل الضرع وشكل الحلمة تاثيرا معنويا في انتاج الحليب اليومي والكلي .2 - بلغ المتوسط العام لانتاج الحليب الجزئي 30 و60 و90 يوم 171.98 و415.10 و656.97كغم على التوالي. وتاثر معنويا (p<0.05) بكل من موسم الادرار وشكل الضرع وشكل فتحة الحلمة.3 - بلغ المتوسط العام لكل من نسبة الدهن والبروتين وكمية اليوريا3.69 % و3.63% و5.48 ملمول/لتر على التوالي وتاثرت هذه المكونات بكل من موسم ومرحلة الادرار, في حين لم يكن للتداخل بين موسم ومرحلة الادرار تاثيرا معنويا لهذه المكونات.4 - اظهر نموذج Wood (1967) غير الخطي ملائمته لانتاج الحليب اذ ينطبق هذا النموذج على القيم الحقيقية لانتاج الحليب منذ بدء الانتاج حتى نهايته والفرو قات بين الانتاج الفعلي والمقدرة بهذا النموذج متقاربة جدا في اي وقت من مرحلة الانتاج. 5 - بلغ الانتاج الابتدائـي للحـليب (الاسبوع الاول a ) المحسوبـة بنموذج Wood (1967) تساوي 33.74 كغم وبزيادة اسبوعية(b) قدرها 0.517 كغم/اسبوع حتى الوصول الى انتاج القمة (65.08كغم), ثم بدا بالانخفاض(c) بمعدل 0.059كغم/ اسبوع وبمثابرة قدرها 74.22% من انتاج القمة.6 - كانت نسبة الدهن والبروتين وكمية اليوريا الابتدائية (انتاج الشهر الاول) المقدرة بنموذج Wood (1967) تساوي %4.023 و%2.89 و5.31 ملمول/لتر على التوالي.7 - تظهر النتائج بان الانتاج الابتدائي (a) يرتبط ارتباطا معنويا وسالبا مع كل من المعيارين b وc والمثابرة وعدد اسابيع القمة ( - 0.48و - 0.68 و- 0.61 و- 0.60 على التوالي) الى انتاج الحليب .8 - تاثرت كل من المثابرة وقمة الانتاج والانتاج الابتدائي (a) لانتاج الحليب ونسبة البروتين معنويا (p<0.05) بموسم الادرار .9 - وجود ارتباط معنوي موجب (p<0.01) بين كمية انتاج الحليب وكمية اليوريواالبروتين والدهن في الحليب, وكان معامل الارتباط بين انتاج الحليب ومكوناته والمعايير الكيموحيوية للدم عالي جدا (p<0.01) وكان الارتباط بين كمية اليوريا في الدم وكمية انتاج الحليب يساوي 0.958 وكمية البروتين في الحـليب (0.698) وكمية اليوريا في الحليب (0.834).10 - بلغ المتوسط العام للارتفاع عند اتصال الرقبة والذنب وطول الجسم المائل وطول الجسم ومحيط الصدر ومحيط البطن والمسافة بين العظمتين الدبوسيتين 121.04, 147.35 , 141.69 ,127.17, 188.69, 200.87, 5.962 سم على التوالي , في حين بلغ المتوسط العام عمق الضرع ومحيطه 39.14 , 17.00 سم على التوالي وكان المتوسط العام لطول الحلمة الامامية والخلفية ومحيط الحلمة 6.87، 5.81 ,7.59 سم على التوالي.11 - اظهر كل من طول الجسم ومحيط الصدر والمسافة بين العظمتين الدبوسيتين وطول الضرع وطول الحلمة الامامية والخلفية ارتباطا موجبا ومعنويا (0.778 - 0.418). 12 - اظهرت جميع انواع الانتاج اعتمادا معنويا (p<0.05) على طول الجسم فقط (انحدار بسيط) وبمعامل تحديد(R2) تراوح بين 21 - 31 % وعند اضافة المسافة بين العظمتين الحرقفتين ومحيط البطن او محيط الصدر(انحدار متعدد) اضافة الى طول الجسم كعوامل مستقلة تراوحت قيمة معامل التحديد(R2) بين 54 - 79 % .13 - تميز انحدار انتاج الحليب والدهن والبروتين الخطي والتربيعي والتكعيبي على طول الجسم بالمعنوية (p<0.05) وكانت قيمة معامل التحديد تتراوح بيـن 23.1 % الى 36.3 %.14 - امتاز الانحدار الاسي الى انتاج الحليب ومكوناته جميعها بالمعنوية (p<0.05) على طول الحلمة الخلفية. | The present study was conducted at Agricultural Research Station, College of Agriculture, University of Basrah and nearby farms. Data of 23 hybrid cows of different ages and season were used. The cows fed wheat bran and reeds as roughages ad libtum . All measurements were done during 1/12/2006 to 31/12/2007. Data included body, udder and teat measurements and partial milk and total milk yield. Milk yield and its content were also included. The result revealed the followings : 1 - Overall mean of daily, total and corrected for 305 day milk yield were 5.80, 1740.97 and 1883.91kg respectively. Entire trait significantly influenced by lactation stage and season. Corrected milk yield for 305 day was influenced by udder shape, teat open and teat shape. However udder and teat shapes did not significantly affect daily and total milk yield. 2 - Mean of partial milk at 30, 60 and 90 days were 171.98, 415.10 and 656.97 kg respectively. They were significantly influenced by season, udder shape and shape of teat open.3 - Mean of fat and protein percentages and were 3.96%, 3.63% and 5.48 mmol/L respectively. Season and stage of lactation had significant effect on these traits, in contrary to interaction effects.4 - Wood’s (1967) model was suitable to milk yield and its component, as it fitted to all early and late yield data beside differences between actual and expected values were very low.5 - Initial milk yield (first week yield, a) calculated by Wood’s (1967) model was 33.74 kg. Milk increased weekly by a rate of 0.517 kg/week to reach peak production (65.08kg), after that decreased in a rate (c) of 0.059 kg/week. Persistency was 74.22% from peak production.6 - Initial fat and protein percentages and urea content (production of 1st month) estimated by Wood’s (1967) model was 4.023%, 2.89% and 5.31 mmol/L respectively.7 - Initial production (a) was significantly correlated with b and c parameter, persistency and number of weeks at peak ( - 0.48, - 0.68, - 0.61 and - 0.60 respectively).8 - Lactation season significantly affected persistency, peak production and initial production (a) for milk yield and protein percentage.9 - There was a significant positive correlation between milk yield and its component (fat, protein and urea). Correlations among milk yield and its component with blood biochemical parameters were high, significant and positive. They were 0.958, 0.698 and 0.834 between blood urea and milk yield or milk protein or milk urea respectively.10 - Overall mean of body height at neck and tail, body length, diagonal body length, chest girth, abdominal girth and distance between pin bones were 121.04, 147.35, 141.69, 127.17, 188.69, 200.87 and 25.96 cm respectively. Mean of udder depth and width were 17.00 and 39.14 cm respectively. Mean and front and rear teat length and teat diameter were 7.59, 6.87 and 5.81 cm respectively.11 - Body length, chest girth, and distance between pin bone, udder length, and length of front and rear teats showed significant correlations ranged 0.418 - 0.778.12 - All type of production regressed significantly on body length (simple regression) with a determinant coefficient (R2) equal to 21 - 31%. When the model included distance between pin bone, abdominal circumference or chest girth (multiple regression) as well as body length as independent variables caused an increase in R2 to be 54 - 79%.13 - Linear, quadratic and cubic regression of milk, fat and protein yield on body length were significant with ranged 23.1 to 36.3%.14 - Expontial regression of milk yield and its components on rear teat length were significant.

Abstract: The study consists of three parts related with patients of CRC as in : 1 - Molecular part 2 - Cytogenetic and molecular genetics part and 3 - Immunological part. In addition of their relatives of clinical and pathological diagnosis, genus, and tumor locations in 150 of Iraqi patients confirmed with CRC collected from Teaching Hospitals of some Iraqi governorates. The percent of male to female (54.66% / 45.43%) and the aged range of study were 16 to 80 years.The first part consist a study of five types of genes related with CRC represented by MLH1, MSH2, APC, K ras, and SMAD7 genes. Some of these genes develop with germline mutations as in MLH 1 exons1 and 15 so as MSH2 genes. While the rest genes related with a sporadic of CRC. All these genes were amplified by conventional polymerase chain reaction (PCR) for study the types of the mutations and their defect of these genes on CRC. The APC exon11and K ras exon1 genes were processed by single strand conformational polymorphism (SSCP) technique. The results of PCR - SSCP technique for two genes represented by appeared three types of bands, which were (AA), (AB) and (AC) bands. The important band was (AB) which constitutes the variability region represented by 84.2% and 44.82% in patients for both genes. The results of patients with mutated bands with high significant differences(P?0.05).compared with healthy control. Patients carried these bands subjected to PCR - Sequences technique. The analysis results for APC gene sequences were substitutions mutations type of single nucleotide polymorphism (SNP) change Cytocine to Thiamin (C>T) at 1972 location of genome. While the analysis results of Kras gene appeared two types of mutations, substitution and frame shifts represented by deletion mutation and so as appeared stop codon mutation.The other genes represented by MLH1, MSH2, and SMAD detected by direct PCR - Sequences. The results analysis of MLH1 gene, exon (1and 15) represented by nucleotides change with high frequencies of substitution mutation through exon1 at different locations of nucleotides. So as frame shift mutation type deletion through exon15. While for MSH2 gene, exon 6 from 6.66% represent by deletion mutation. A spread study for SMAD7 gene, exon4 were done through 30 patients average age 52 years (male to female 56.7% : 53.3%) confirmed colorectal adenocarcinoma compared with 15 healthy. A mutation of 20% of (SNPs) were identified.The second part which represented standard conventional cytogenetic and molecular genetics were done through peripheral blood culture (PBC). The study showed increasing of mitotic index (MI) in patients with significant differences (P?0.05) compared with healthy controls indicating for increasing of proliferation of the lymphocytes division. The study showed some chromosomal aberrations with significant, the majority of aberrations were higher significant in dwarf and aneuploidy chromosomes so as elongation chromosomes revealed significant differences (P?0.05) between patients and healthy. The appeared of Polyploidy chromosomes, with significant differences (P?0.05), that’s seem to be good indicator for cancer disease. The molecular genetics alterations which dependent on cytogenetic investigation were done through deletion of long arm of 5 chromosome which APC gene is located. The test elucidates the importance of inactivation (deletion) of APC gene in elicited of colorectal cancer. The study of 20 patients, were investigated by fluorescent in situ hybridization (FISH). The results showed 60% patients which have 5q arm deletion of (+). And (5%) have two deletions (++) which was a male aged less than 60 years. The results analysis explained significant differences (p?0.01) between studied group and deletion (+) of arm 5q. A correlation represented by significant differences (p?0.01)through dysfunction of APC gene deletion of 5q were found with high percent among sporadic adenocarcinoma 58.3%, with less in attenuated familial adenomatous polyposis (AFAP) and familial adenomatous polyposis (FAP) represented by the (25%), and (16.7) respectively.The last part related with immunological test were done by detection of tumor markers represented by Carcino Embryonic Antigen (CEA), IL - 33 and IL - 31.These markers represented a confirmative indicators for colorectal adenocarcinoma which were tested by ELISA technique in (79) patients confirmed with CRC of male and female. These tumor markers in tested with patients showed high significant differences (p?0.01) compared with healthy controls. Another immunological test for detection of Human papilloma virus (HPV) type 18 E6 gene expression in CRC by Immune histochemistry technique (IHC) in 71 (FFPET) of CRC. The results of this study showed there were a correlations between patients and adenocarcinoma positive HPV18 E6 infection 43.7% of patients infected with virus, with high significant differences (p?0.01) and higher infection in female (67.7%) than male (32.3%). So as a correlations were found between HPV18 E6 expression and tumor tissue locations, explained by high percentage 45.2% in rectum.While a tumor marker was (CD8+) was used to determine its role in human immune surveillance in tumor regions of CRC so as with regions which were infected by HPV. The study revealed high significant differences between healthy control and studied groups which have (CD8+) positive infiltration in tumor origins through detection by (IHC).In conclusions diagnosis of CRC can be detected by special genes like K ras gene through professional PCR processing, compact with tumor markers, so as Fluorescent in situ hybridization (FISH) technique provides a precise method which can be used for detection of alteration of molecular and cytogenetic related with CRC.

For the purpose of this study, preliminary experiments were carried out to find the most effective doses of plant extracts and it was found that these doses were 100, 150, 250 mg/kg body weight for, tomato, celery and gordeen roket extracts respectively. Compared with the control, the oxidative stress induced by H2O2 caused significant increase in total count of leukocytes, neutrophilis, acidophils, basophilis, monocytes and lymphocytes, and in the level of blood sugar (glucose), Total cholesterol, triglycerides(TG), low density lipoprotein (LDL - C), very low density lipoprotein (VLDL - C) atherogenic index (First and second), Urea, Malondialdehyde (MDA) and percentages of dead sperms and abnormal sperms.This stress showed as well, a significant decrease in body weight, Hb, PCV, high density lipoprotein (HDL - C), total protein, Albumin, glutathione, sperm total number, percentages of motile and normal sperms, testes weight, prostate and epididymis weights and seminiferous tubules diameter. This treatments, however, showed no significant variations in activity levels of ALT and AST enzymes and seminal vesicle weight.2. In comparison with treatment (2) (rats treated only with H2O2) , the treatment of rats, under oxidative stress with celery extract showed a significant decrease in total count of leukocytes neutrophils, lymphocytes and in blood suger, cholesterol TG atherogenic (First index) blood urea, MDA, Percentage of dead and abnormal sperms and seminal vesicle weight.This treatment showed also a significant increase in level of Hb, PCV., body weight, total protein, Albumin glutathione, number of sperm, percentages of motile and normal sperms, testis weight, epididymis weight and prostate weight. However no significant variations were detected in levels of acidophils, basophils, monocytes, HDL - C, atherogenic (second index) and seminituferous tubules diameter.3. Treatment of Eruca extract and compared with the treatment of H2O2, showed significant decrease in lymphocytes number and in the level of blood sugar, cholesterol (T.G), LDL - C, VLDL - C and atherogenic (first and second index) blood urea, MDA, percentage of dead and abnormal sperms and seminal vesicles weight. The treatment showed also a significant increase in body weight, numbers of acidophils, monocytes, basophils, HDL total sperm numbers, percentages of motil and normal sperms and weights of testis, epididymis and prostate.However, no significant variations were observed in Hb, PCV, Leukocyte total counts, neutrophil, albumin, ATL and AST and seminiferous tubule diameter.4. The treatment of tomato extract showed similar results to that of celary and Eruca extracts except, a significant increase in AST enzyme and seminiferous tubules diameter was detected in case of tomato extract (i.e not seen in case of other plant extracts).5. Compare with H2O2 treatment, rats under oxidative stress treated with vitamin E (500 mg/kg b.w) showed significant decrease in lymphocyte numbers, Total cholesterol, TG, LDL - C, VLDL - C, atherogenic (first and second index), urea, MDA and percentages of dead and abnormal sperms.At the same time the vitamin E treatment showed a significant increase in body weight , Hb, neutrophils, acidophils , monocytes, total protein, glutathione, total sperms number, percentage of motile and normal sperms, and weights of testis, epididymis, and prostate.However, this treatment (compared with H2O2 treatment) showed no significant difference in the level of PCV total count of leukocytes, basophils, blood sugar, HDL - C, albumin, ALT, AST, seminal vesicle weight and seminiferous tubules diameter 6. The most important histological changes exhibited by the animals under oxidative stress were apparent interstitial degeneration, necrosis of supporting and spermagonial cells, separation of these cells in semicyclic manner from the basement membrane, decrease in spermatids (spermatoblasts) and mature sperms in seminiferrous tubule cavity.These effects (due to H2O2 treatment) were decreased to minimal levels when the H2O2 treated rats exposed to plant extracts and vitamin E, used in this study.7. Animals treated with plant extract or vitamin E alone (i.e not treated before with H2O2) showed no clear difference from the control animal (without any treatment) though some improvement was observed in some physiological, biochemical and histological characters of blood and male reproductive system of the studied animals The above results revealed that the plant extract used in this study play an important role in decreasing the harmful effect of the free radicals in the animals (rats) studied.
