Abstract: This study included isolation and identification Candida albicans from different sources ,We obtained (26) isolates of Candida albicans of the total (115) samples which included (60) clinical samples of children who suffer from inflammatory in the Area of diapers in the children Protection Teaching - Hospital in The Medical City hospital and Alalweia hospital in Baghdad ؛(50) soil samples from Nurseries and gardens and corrals , )5) dried milk samples in the period between January2012to April2012 . We obtained (20) isolates C. albicans from the clinical samples; (5) isolates from soil samples and only one isolate from the dried milk samples.All isolates have been cultured on a differential mediums to diagnosis and identify C. albicans from the other species . Biggy agar, Candida medium, Candida BCG ,Corn meal agar involved in the isolation and identification of the C. albicans. Candida diagnosed based on the phenotypic characteristics of the developing colonies.All isolates was involved to microscopic examinations for the purpose of examining the phenotypic characteristics of C. albicans cells as well as to examine the germination tube formation, The diagnosis also includes the use of biochemical methods such as fermentation and assimilation behavior of Candida , API20cAUX System and ability of Candida to produce Proteinase in Skim milk agar and Blood agar.The second axis of the current study included investigating about white - opaque phenotypic switching, (20) clinical isolates (5) soil isolates (1) milk isolate showed ability to transition from white to opaque ,also we found the results of phenotypic transition ratios of the colonies grown on modified Lee’s mediums by adding Glucose or N - acetyl glucosamine in different incubation conditions involved the aerobic condition and presence of CO2, at (25)°c and (37)°c during (1,2,5,and 10) days. the results of transition ratios and the overlap between the factors compared statistically. the study found that the ratio reached a maximum at physiological temperature (37 o C) in the presence of N - acetylglucoseamine and 1% CO2 . in the aerobic condition at (37o)C with presence of N - acetylglucoseamine the results show a significantdifference at the duration between the 1st and 2nd day, and no significant difference on the dried milk isolate . The results of 5th and 10th days do not show significant difference in any isolates which due to the saturation condition when cell do not affect a change in the metabolic activity, so it dispense with N - acetylglucoseamine who induce the swathing only for a short duration.Further study the morphology of C. albicans with transmission electron microscope (TEM) and the virulence of the white and opaque colony of C. albicans. The histopathological study of the liver, the spleen and the kidneys of injected mice (Balb/c) showed several changes in the tissues, two concentration of yeast suspension (1.5 x 108) cells/ml and (3x 108) cells/ml prepared by McFarland method are used, the injection with the white the phenotypic (1.5 x 108) showed necrosis in the renal tubule and mononucleosis filtration in the tissue of the kidney, also simple filtration to the inflammatory cell in liver and a replication and necrosis in the lymphatic tissue of the spleen .The white the phenotypic (3x 108) showed a necrosis in the lymphatic tissue of the spleen and a filtrationto the Megakaryocytes, acute necrosis in liver cells and a filtration to the inflammation near the hepatic portal veins and a acute renal failure combined with mononucleosis filtration in the kidney ,On the other hand the opaque cells injection (3x108) cells/ml has caused a simple necrosis in tissues of the liver, spleen and the kidneys while the opaque cells (1.5 x108) cells/ml has no effective virulence and the tissue diagnosed as a normal.
