Abstract:
The present was carried out during the period of November 2014 to April 2015 in Al - Hilla City. This study included 100 samples which were collected from patients at ages ranged between 4 to 75 years old whom were suffering from dental caries and periodontal diseases ; (50) samples were taken from patients infected with dental caries (23 male and 27 female) and (50) samples taken from patients with periodontal diseases (15 male and 35 female), who have submitted to a college of Dentistry in Babylon university and special center of Dentistry. Thirty samples were taken from apparently healthy persons and considered as control.The isolation and identification of S. mutans was performed by Mitis Salivarius Bacitracin agar as a selective medium and biochemical tests as well as API 20 strep, then investigated the virulence factors and show the effect of some antibiotics on bacterial isolates, and the identification of E. gingivalis parasite was by a wet preparation method.The obtained results showed that S. mutans was with a higher existence in patients with dental caries, with lower existence in the patients with periodontal diseases and it was found in healthy persons (30%, 18%, and 0%) respectively. Entamoeba gingivalis was higher in patients with periodontal diseases than patient with dental caries and less in healthy individuals (62%, 24%, and 6%) respectively. The presence of these microorganisms was related to the type of dental disease (p=0. 009), gender (p=0. 009), age (p=0. 008) and risk factors (diabetes, smoking and hypertension) (p=0.037).The effect of some antibiotics on S.mutans was also tested, and the obtained results showed that all isolates were with high resistance to Erythromycin (70.8%) and (66.7%) were resistant to Doxycycline. Some isolates showed low level of resistance to Vancomycin (54.2%), CefatoximeII(33.4%) and Clindamycin (25%), while these isolates were with high susceptibility to Chloramphenicol (100%) and Ofloxacin (83.4%).Finally, some virulence factors of S. mutans were studied and it was seen that the (62.5%) of S. mutans isolates produced α - hemolysis and (8.3%) produced β - hemolysis but (29.2%) of isolates non - hemolysis, but (58.3%) of isolates were made capsule and (100 %) were able to synthesize glucan, (83.3%) of S. mutans isolates were able to produce mutacin and (45.83%) of isolates were found to produce dextranase enzyme, while (84%) were able to biofilm formation .In addition, all isolates in this study were able to adhere to oral epithelial cells.
