Abstract: Oxidative stress plays an important role in the pathogenesis of cardiovascular alterations observed in diabetic patients and that hyperglycemia is the causal link between diabetes and increased oxidative stress. Evidence suggests that Ser326Cys, a genetic polymorphism of (8 - oxoguanine glycosylase 1 OGG1), is associated with coronary artery lesions in patients with diabetes mellitus type 1; however, the underlying mechanism is unclear.Aims of the study : - Compare the genetic distribution of genetic polymorphism genotype 8 - oxoguanine glycosylase1 (OGG1) in Type1 diabetic with number of healthy people; Sequencing of amplified product of OGG1 gene for the studies group; Evaluation of some clinical characterized on lipid profile, FBG, HbA1c, and OX - LDL. Finally study the relations between OGG1 gene polymorphism and clinical characters.Material and methods : - The study involved a total (100) subjects including (60) people are suffering from diabetes Type1 there’s aged between (10 - 40 years) and (40) non - diabetic controls with aged (10 - 40) years. Kits based on enzymatic methods measured serum concentrations of glucose and lipid profile. Oxd - LDL was measured by using standard enzyme linked immunosorbent assay (ELISA) method, Hemoglobin (A1C) by kit based on non - enzymatic method also were measured the total DNA by purification kit , amplified OGG1 in DNA by used PCR kit, and determination of intraspecies variation for OGG1 by used restrictions enzyme Fnu4HI.IIResults : - Genetic analysis for OGG1 genetic polymorphism show that the genotype (C/C) and genetic pattern hybrid (C/G) in patients group (26.67%), (73.33%) compared with the control group (100 %), (0.00%) and the difference was significant (P≤0.01), while (G/G) genotype frequency was found no results, so there was not significantly different (p>0.05) neither in the diabetes mellitus type1 nor controls (0.00%). Genotyping data Sequencing of OGG1 gene from (healthy and patient) revealed 100% compatibility with Homo sapiens for OGG1 type 1 gene, and ether them (patient) appeared 99% compatibility with Homo sapiens for OGG1 type 1 gene.Oxidized Low Density Lipoprotein (Oxd - LDL) level in patients were significant when compare with healthy controls.Conclusion : - our study revealed that the OGG1 could have an important role in the development of type1 diabetes; also, ROS have an essential function that is impact on fat indicators was shown in the Oxd - LDL.
