Abstract:
يمثل هذا البحث دراسة اولية لتقييم تاثير المستخلصات الخام لجمار نخيل التمر صنف الزهدي Phoenix dactylifera cultivar. Zahdi في نمو اثنين من الخطوط الخلوية السرطانية هما خط خلايا سرطان العضلة البشري RD وخط خلايا سرطان الخلايا الدبقية البشرية ANG وخط واحد من الخلايا الطبيعية لجنين الجرذ REF والخط المتحول وراثيا VERO فضلا عن دراسة تاثيرها في انقسام الخلايا اللمفاوية البشرية . تم تحضير ثلاثة مستخلصات خام لجمار النخيل وهي مائي(باستعمال الماء المقطر مزدوج التقطير ) وكحولي (باستعمال كحول الايثانول 70% )والعصير الخام (وذلك بعصر الجمار واستعماله مباشرة ). واستعملت اربعة تراكيز مختلفة محضرة بشكل ثنائي من هذه المستخلصات وهي ( 1250، 2500 ، 5000، 10000) مكغم/مل بالنسبة للمستخلص المائي والكحولي ، و(1% ،5% ،10% ، 30% )بالنسبة للعصير ، وتم اختبارها لكل من خطوط الخلايا السرطانية والخط المتحول وراثيا بمعدل ثلاث مكررات لكل تركيز ولمدة معاملة (24، 48 ، 72 )ساعة باستثناء الخلايا الطبيعية التي كانت مدة معاملتها 72 ساعة فقط . وقد اظهرت النتائج وجود تاثير سمي معنوي واضح للمستخلصات الخام في نمو الخلايا السرطانية لثلاث مدد من التعريض ، ووجد ان التاثير التثبيطي للمستخلصات يعتمد على التركيز حيث كانت السمية تزداد بزيادة التركيز وعلى وقت التعريض لبعض التراكيز . لقد اختلفت افضلية المستخلص الاكفا في التاثير السمي على الخطوط الخلوية السرطانية باختلاف نوع المستخلص والتركيز والخط الخلوي السرطاني. اذ كان العصير الخام اكفا المستخلصات الخام تاثيرا في الخط الخلوي السرطاني ANG حيث تراوحت نسب التثبيط لهذا المستخلص من 69.7% للتركيز 1% الى 81.5% للتركيز 30% لمدة التعريض 24 ساعة ، اما في 72 ساعة فقد تراوحت النسب بين (73.8% - 83%) للتراكيز (1% - 30%). اما في الخط الخلوي السرطاني RD والخط المتحول وراثيا VERO فقد كان المستخلص المائي هو الاكفا حيث كانت اعلى نسب للتثبيط عند التركيز 10000مكغم/مل وكانت النسب تنخفض بانخفاض التركيز حيث بلغت النسب (90.7 %، 89%، 88.8% ) (جدول4 - 8) للخط RD و(92.9%، 93.7% ، 91.8% )(جدول 4 - 2) للخط الخلوي VERO لمدد التعريض (24، 48 ، 72 ) ساعة على التوالي اي ان مدد التعريض لم يكن لها تاثير معنوي على نسب التثبيط. عند استعمال المستخلصات الخام بديلا عن المادة المشطرة PHA ، لم يؤد الى تحفيز انقسام الخلايا اللمفية اي انه لايعمل كبديل عن ال PHA . ولكن عند استخدامه كبديل عن الكولجسين لوحظ انه يعمل على ايقاف انقسام الخلايا في مرحلة ال Metaphase مما يدل على ان المستخلصات اظهرت دورا مشابها لعمل الكولجسين ولكن بكفاءة اقل . | This work represented a preliminary study of the effect of crude extracts of date palm pith (Phoenix dactylifera cultivar. Zahdi) on the growing of two cancer cell lines(Human Rabdomyosarcoma - RD) , (Glioblastoma - ANG) and one normal cell line (Rat Embryo fibroblast - REF) and the transformed cell line (VERO). In addition, it involves the study of their effects on the division of the human lymphoid cells.Three crude extracts of date palm pith were prepared which is aquous (using double distilled water), alcohol (using ethanol 70%) and the crude juice(squeezing the pith and use it directly).Four different binary concentrations have been prepared from crude extracts that are concentrations(1250,2500,5000,10000) µg/ml for each aquous and alcoholic, (1%, 5%, 10% 30%)for the crude juice. Each cancerous cell line and the transformed cell line was tested with an average of three repeaters for each concentration and for the treatment periods of (24 ,48 ,72) hours except for the natural cells whose treatment period was only 72 hours . The results show existence of clear significant cytotoxic effect for crude extracts on growth of cancer cells within the three periods of time of exposure .It was found that the cytotoxic effect for crude extracts and depended on concentration and time period of exposure. It is noticed that the cytotoxic effect increased according to concentration .The Effectiveness of more extract in terms of cytotoxicity on cancer cell lines has been varied depending on types of extract , cancer cell line and concentration.the crude juice was found to be the most effective especially on the cell line (ANG) The inhibition rate for this extract were (69.7%) for the concentration 1% to 81.5 for concentration 30% for exposures period 24 hour , while in 72 hour The inhibition rate were (73.8% - 83% ) for concentrations (1% - 3%). While on RD cell line and the transformed cell line (VERO) the aquous extract was more effective , the highest inhibition rate was for the concentration 10000 µg/ml the inhibition rates was increased according to concentration .It was (88.8% , 89% , 90.7%)for RD cell line and (91.8%, 93.7% , 91.8%) for VERO cell line for exposure periods (24, 48, 72) hour respectively. When we use the crude extracts as an alternative for the PHA , it is found that it doesn’t have the ability to motivate the cells division of the lymphoid cells , (i.e. it can't be as alternative for the PHA ). When using it as alternative for colchicin. It is noticed that it works on stopping the division in the tropical phase this is improve that these extracts show the same work like colchicin but less than it.
